Auxiny zajišťují základní procesy v rostlině zejména dělení buněk a elongaci. Ve vyšších koncentracích však mohou být pro rostlinu škodlivé, proto jsou ukládány většinou v neaktivní formě, kterou představují konjugáty. V případě konjugátů s aminokyselinami jsou důležitými enzymy auxin-aminokyselinové hydrolasy, které štěpí amidovou vazbu aminokyseliny konjugované auxinem a uvolňují tak volnou aktivní formu, což je jednou z možností, jak je hladina auxinů v rostlině regulována a je tak zajištěna homeostáza. Funkce těchto hydrolas byla charakterizována u mnoha rostlin (Arabidopsis thaliana, Oryza sativa), nicméně u ječmene (Hordeum vulgare) doposud zkoumány nebyly. Podle provedené transkriptomické analýzy u ječmene se exprese těchto genů mění během raného vývoje nodálních kořenů, což by mohlo mít zásadní vliv při zakládání kořenů a tím i na reakci rostliny ke stresu suchem, který je aktuálním problémem v zemědělství. V rámci diplomové práce byly nalezeny geny pro domnělé auxin aminokyselinové hydrolasy u Hordeum vulgare a sekvence tří z nich byly klonovány do dvou vektorů pro bakteriální expresní systém. Následně byly vektory transformovány do expresních buněk Escherichia coli a byla provedena optimalizace podmínek exprese vybraných auxin amidohydrolas Vzhledem k ukládání proteinu v inkluzních tělískách byly aplikovány různé podmínky s cílem získat protein v solubilní frakci, a purifikovat jej pomocí afinitní chromatografie.Auxins ensure key plant processes, especially cell division and elongation. However, at higher concentrations, auxins can be harmful to the plant, therefore they are mostly stored in an inactive form represented by conjugates. In the case of conjugates with amino acids, auxin-amino acid hydrolase enzymes are important, which cleave the amide bond of the auxin-conjugated amino acid and thus release the free active form of auxin. This is one of the ways how the level of auxins is regulated. The function of these hydrolases has been characterized in many plants (Arabidopsis thaliana, Oryza sativa), however, they have not been studied in barley (Hordeum vulgare) yet. According to the performed transcriptomic analysis in barley, the expression of these genes have changed in the early development of nodal roots, which could have a fundamental effect on root establishment and thus on the plant's response to drought stress, which is very discussed problem of agriculture. In the diploma thesis, the genes for putative auxin amino acid hydrolases of Hordeum vulgare were found and the sequences of three of them were cloned into two vectors for the bacterial expression system. Subsequently, the vectors were transformed into Escherichia coli expression cells and the expression conditions of selected auxin amidohydrolase were optimized. Due to the deposition of the protein in the inclusion bodies, various conditions were applied to obtain the protein in the soluble fraction. Finally proteins were purified by affinity chromatography.
