Bakteriální a houbová společenstva ovlivňují rovnováhu všech ekosystémů. Jsou jedním z klíčových faktorů pro správný růst a výživu rostlin. Pro detekci mikroorganismů a studium mikrobiálních společenstev se v současnosti uplatňuje hlavně metoda PCR a 'high throughput sequencing'. Cílem této práce byla identifikace druhů bakterií a hub vyskytujících se na povrchu poškozených plodů meruněk pomocí PCR amplifikace variabilních oblastí 16S rDNA bakterií a ITS oblastí u hub a identifikace mikroorganismů na základě analýzy sekvencí získaných Sangerovým sekvenováním a sekvenováním nové generace. Pomocí konvenčního přístupu, který zahrnoval Sangerovo sekvenování se na povrchu plodu meruňky nepodařilo identifikovat žádný fytopatogenní mikroorganismus. Pomocí sekvenování nové generace bylo na povrchu plodů meruněk s poškozením a bez poškození detekováno devět rodů hub, ale nebyl prokázán jejich přímý vliv na poškození plodu.Bacterial and fungal communities affect the balance of all ecosystems. They are one of the key factors for good plant growth and nutrition. Nowadays, PCR and 'high throughput sequencing' are mainly used to detect microorganisms and study microbial communities. The aim of this work was to identify species of bacteria and fungi occurring on the surface of damaged apricot fruits by PCR amplification of variable regions of 16S rDNA bacteria and ITS regions of fungi, and identification of microorganisms based on sequence analysis obtained by Sanger sequencing and next generation sequencing. Using a conventional approach using Sanger sequencing, no phytopathogenic microorganism could be identified on the surface of the apricot fruit. Using next-generation sequencing, nine genera of fungi were detected on the surface of apricot fruits with and without damage, but their direct effect on fruit damage was not proven.
