Předkládaná bakalářská práce si klade za cíl zkoumání interakce mezi signalizací fototropinů a tolerancí k abiotickým stresům, konkrétně k zasolení a u modelové rostliny huseníček rolní (Arabidopsis thaliiana (L) Heynh.). Teoretická část shrnuje dosavadní znalosti o rostlinných fototropických reakcích a reakcích k zasolení a zároveň se také věnuje rostlinným hormonům auxinu a kyselině abscisové, které se na těchto dějích podílejí. Experimentální část se pak zabývá zkoumáním vlivu fototropinů na toleranci k zasolení a vlivu zasolení, společně s inhibicí syntézy kyseliny abscisové, na fototropické reakce hypokotylů Arabidpsis v podmínkách in vitro. Výsledky této práce vedou k závěrům, že stres ze zasolení inhibuje v hypokotylech Arabidopsis fototropickou reakci, a to alespoň z části nezávisle na inhibici elongace hypokotylu a rovněž částečně vlivem kyseliny abscisové, která může být negativním regulátorem fototropismu i při nestresových podmínkách. Výsledky také naznačují, že by se fototropiny mohly pozitivně podílet na toleranci hypokotylů Arabidopsis k zasolení.This bachelor thesis aims to investigate the interaction between phototropin signaling and plant abiotic stress tolerance specifically to salinity and in the model plant Arabidopsis thaliana (L) Heynh. The theoretical part summarizes existing knowledge about plant phototropic responses and responses to salinity, and simultaneously it is focused to plant hormones auxin and abscisic acid, involved in these processes. The experimental part examines the effect of phototropins on salt stress tolerance and the effect of salt stress along with inhibition of abscisic acid synthesis on phototropic reactions of Arabidopsis hypocotyls in in vitro conditions. The results of this thesis lead to conclusion that salt stress inhibits the phototropic reactions of Arabidopsis hypocotyls at least in part independently on inhibition of hypocotyl elongation and also partly due to abscisic acid, which may be a negative regulator of phototropism even under non stress conditions. The results also suggests that phototropins may be positively involved in the tolerance of Arabidopsis hypocotyls to salt stress.
