Tato práce je zaměřena na monitoring transkripční aktivity vybraných receptorů pod vlivem syntetických derivátů přírodních látek. V první části práce je teoreticky popsána rodina nukleárních receptorů, a signální dráhy obou testovaných receptorů (aryluhlovodíkového AhR a glukokortikoidního GR), společně s jejich strukturou a funkcí. Tyto receptory jsou ligandem aktivované transkripční faktory. V praktické části je za pomoci MTT testu zjišťována cytotoxicita syntetických látek a následně jejich schopnost aktivovat nebo inhibovat transkripční aktivitu receptorů, což je měřeno pomocí metody Gene reporter assay. Zatímco pozitivní kontrola dexamethason (DEX) významně aktivoval GR, žádná z testovaných látek GR neaktivovala. Signifikantní inhibice aktivovaného GR byla zaznamenána pro KAC 36 (100 M). V případě AhR byla zaznamenána signifikantní aktivace pro nejvyšší koncentrace testovaných látek, které dosahovaly méně než 10% aktivace pozitivní kontrolou dioxinem TCDD. Výjimku tvořila látka KAC 36 (50 M), která dosahovala asi 33% aktivace pozitivní kontroly, TCDD. Všechny testované látky vykázaly statisticky významný inhibiční účinek pro dioxinem aktivovaný AhR. I přes nízkou schopnost látky KAC 36 (100 M) inhibovat aktivitu luciferázy (cca o 10 %), lze soudit, že testované látky působí jako parciální agonisté aryluhlovodíkového receptoru.This work is focused on transcription activity monitoring of chosen receptors influenced by synthetic derivatives of natural compounds. In the first part of this work, there is described superfamily of nuclear receptors and signaling pathways of both receptors (aryl hydrocarbon AhR and glucocorticoid GR) including their structure and function. These receptors are ligand-activated transcription factors. The practice part is dedicated to the detection of cytotoxic effects caused by synthetic compounds using the MTT test, followed by detection of their ability to activate or inhibit transcription activity of receptors measured by Gene reporter assay method. While positive control dexamethason (DEX) significantly activated GR, none of tested compounds did. Significant inhibition of activated GR was measured for KAC 36 (100 M). In the case of AhR, significant activation was detected for the highest concentration of tested compounds with less than 10% activation by positive control dioxin - TCDD. The exception was KAC 36 (50 M), which reached approx. 33% activation by positive control TCDD. All tested compounds demonstrated statistically significant inhibitory effect for AhR activated by dioxin. Despite of low ability of compound KAC 36 (100 M) to inhibit luciferase activity (approx. for 10 %), it is possible to deduce that the tested compounds work as partial agonist of aryl hydrocarbon receptor.
