Pšenice setá (Triticum aestivum L.) patří z hlediska výživy mezi jednu z nejdůležitějších plodin světa. V současnosti však její produkce nedokáže plně pokrýt poptávku stále se rozrůstající lidské populace a je ohrožována měnícími se klimatickými podmínkami. Osevné plochy není možné neomezeně rozšiřovat, proto je nutné šlechtit kultivary odolné vůči biotickým a abiotickým stresům a se stabilním a vysokým výnosem. K tomu je potřeba dostatek kvalitních genetických map s markery v těsné vazbě s lokusy pro agronomicky významné znaky. Velikost a komplexita genomu pšenice je hlavní obstrukcí v tomto úsilí, proto jsou pro zjednodušení mapování používány příbuzné druhy se sníženou ploidií. V případě genomu A pšenice to nejčastěji bývá T. monococcum. Cílem předložené práce bylo přispět k mapování agronomicky důležitých znaků testováním vhodnosti IRAP markerů pro mapování u pšenice. Práce se skládá ze dvou částí, teoretické a praktické. Teoretická část práce je zaměřena na přehled mapovacích a markerovacích technik a výnosových prvků pšenice. Praktická část práce je zaměřena na testování vhodnosti IRAP markerů pro zahuštění genetické mapy a mapování výnosu a prvků výnosu u T. monococcum. Celkem bylo analyzováno 108 IRAP primerů na 89 liniích RIL mapovací populace z křížení planého T. monococcum ssp. aegilopoides G3116 a kultivovaného T. monococcum ssp. monococcum DV92. Polymorfní DNA fragmenty mezi rodiči mapovací populace byly pozorovány u 20 % z nich. Počet polymorfních DNA fragmentů na jeden IRAP se pohyboval v rozmezí 1 až 5. Celkově bylo získáno 40 IRAP markerů, které byly použity k zahuštění mapy T. monococcum a byl u nich hodnocen potenciál pro mapování prvků výnosu.Bread wheat (Triticum aestivum L.) belongs in term of nutrition among one of the most important crops in the world. Recently, its production does not fully cover requirements of groowing human population and above that is compromised by changing climatic conditions. Also, extensiont of aralbe land is limited. Breeding cultivars resistant to biotic and abiotic stress and with stable high yield is the main solution to these problems. For fast and efficiens breeding high-quality genetic maps with markers tightly linked to agronomically important traits are crucial. Hovewer, size and complexity of wheat genome is the main obstruction in this efforts. One of a ways to facilitate the mapping is using wheat relatives with lower ploidies. In case of wheat genome A the T. monococcum it is the most common substitute. The ain aim of the proposed work was to contribute to the T. monococcum mapping by testing suitability of the IRAP markers for wheat mapping. The work is composed of two parts, theoretical and practical. The theoretic part of the work is targeted to reviewing of present state of wheat markers and yield traits mappings. The practical part of this work is focused to testing of the IRAP markers for implementation to wheat genetic maps and for mapping of yield traits in T. monococcum. 108 IRAP primers were analysed on 89 lines of RIL mapping population from the cross of wild T. monococcum ssp. aegilopoides G3116 and cultivated T. monococcum ssp. monococcum DV92. 20 % of them showed polymorphic DNA fragments between parental lines of the mapping population. In general 1 to 5 polymorphic DNA fragments per primer were observed. Totally 40 IRAP markers were acquired and mapped to the T. monococcum map and their value for yiedl traits mappig was evaluated.
