Diplomová práce se zabývá purifikací proteinů interagujících se steroidními látkami. V teoretické části jsou shrnuty informace o steroidních hormonech, jejich významu v rozvoji nádorových onemocnění, estrogenových receptorech, proteinech a afinitní chromatografii. V experimentální části je popsána extrakce proteinů z nádorové linie MCF-7 a purifikace proteinů pomocí afinitní chromatografie. Afinitní chromatografie je separační technika pro čištění specifických molekul nebo skupin molekul z komplexní směsi s využitím specifické vazebné reverzibilní interakce mezi molekulami. Purifikované proteiny byly dále zpracovány proteomickými metodami.The diploma thesis deals with purification of proteins that are interacting with steroids. The theoretical part summarizes information about steroid hormones, their importance in development of cancer, estrogen receptors, proteins and affinity chromatography. In the experimental part, the extraction of proteins from the MCF-7 tumor line and purification of proteins by affinity chromatography is described. Affinity chromatography is a separation technique for purifying specific molecules or groups of molecules from a complex mixture using a specific binding reversible interaction between molecules. Purified proteins were further processed by proteomic methods.
