Kvetení u pšenice seté (Triticum aestivum L.) je řízeno geny vernalizační a fotoperiodické dráhy a geny ranosti per se. Porozumění procesu kvetení může přispět k vyšlechtění nových kultivarů se zvýšenými výnosy. Hlavním genem vernalizační dráhy je VRN1, který se u pšenice seté vyskytuje ve třech homeologních variantách: VRN-A1, VRN-B1 a VRN-D1. U ozimých odrůd pšenice jsou všechny tři homeologní geny v recesivní formě a jejich exprese je indukována delším působením nízkých nenulových teplot - vernalizací. V rámci praktické části této bakalářské práce byly pěstovány dvě skupiny rostlin složených ze čtyř linií odvozených od odrůdy Triple Dirk - TDB, TDC, TDD a TDE. Linie TDC měla všechny geny vrn-1 v recesivní formě, zatímco ostatní linie měly vždy jeden VRN1 gen v dominantní formě. Jedna skupina rostlin byla vernalizována a druhá byla pěstována bez působení vernalizace. Z rostlin byly odebrány vzorky ve čtyřech časových odběrech, u kterých byla stanovena exprese jednotlivých alel genu VRN1. Dále byla u rostlin provedena fenotypová analýza (doba metání), která byla porovnána s výsledky exprese. Poslední částí bakalářské práce bylo stanovit poměr premRNA a mRNA u recesivní a dominantní alely VRN-A1. Získaná data ukazují, že nejsilnější alelou vernalizační dráhy je Vrn-A1a, pak Vrn-D1 a Vrn-B1. Alely Vrn 1 také částečně aktivují expresi vrn-1. U nevernalizovaných rostlin nesoucí alelu vrn-1 je přítomno více premRNA, zatímco u rostlin nesoucí alelu Vrn-A1a je více mRNA. Po vernalizaci se poměr premRNA:mRNA VRN-A1 vyrovnal.Flowering in bread wheat (Triticum aestivum L.) is controlled by vernalization and photoperiodic pathway genes and earliness per se genes. Understanding of the flowering process can contribute to the breeding of new cultivars with increased yields. The main gene of the vernalization pathway is VRN1, which occurs in three homoeologous variants in bread wheat: VRN-A1, VRN-B1 and VRN-D1. In winter wheat varieties, all three homoeologous genes are in recessive form and their expression is induced by prolonged exposure to low but non-freezing temperatures - vernalization. In the experimental part of this bachelor thesis, two groups of plants consisting of four lines derived from the Triple Dirk variety - TDB, TDC, TDD and TDE were grown. The TDC line has all vrn-1 genes in the recessive form, while the other lines always have one VRN1 gene in the dominant form. One group of plants was vernalized, and the other was grown without vernalization. The plants were sampled at four time points and the expression of the individual alleles of the VRN1 gene was determined. Furthermore, a phenotypic analysis (heading time) was performed, which was compared with the expression results. The last part of the bachelor thesis was to determine the ratio of premRNA and mRNA in the recessive and dominant allele VRN-A1. The experimental data shows the strongest allele of the vernalization pathway is Vrn-A1a, then Vrn-D1 and Vrn-B1. The Vrn-1 alleles also partially activate vrn-1 expression. In non-vernalized plants carrying the vrn-1 allele, more premRNA is present, while in plants carrying the Vrn-A1a allele, more mRNA is present. After vernalization, the premRNA:mRNA VRN-A1 ratio equilibrated.
