Cílem této diplomové práce bylo charakterizovat funkci aldehyddehydrogenas z rodiny 2 v Hordeum vulgare a nukleosidas z rodiny 2 a 3 v Zea mays. V rámci teoretické části této práce byla vypracována literární rešerše zabývající se problematikou aldehyddehydrogenas, jejich výskytu a funkce v rostlinách i živočiších. Dále byl popsán metabolismus purinových a pyrimidinových bází a funkce nukleosidas v rostlinách. V experimentální části je obsaženo vyhledávání genů ALDH v genomu ječmene ve webových databázích GenBank, Phytozome a UniProt s užitím softwaru BLAST, kdy bylo nalezeno 15 genů. Dále byla tato část zaměřena na klonování, expresi, purifikaci proteinu ALDH2 z ječmene. Pro tento enzym bylo stanoveno pH optimum a byla proměřena substrátová specifita. Na základě výsledků bylo vybráno šest substrátů, u kterých byly stanoveny saturační křivky a z nich dopočítány kinetické parametry Km, Vlim a relativní poměr Vlim/Km. Nakonec byly exprimovány a purifikovány enzymy NRH2b a NRH3 z kukuřice. U těchto proteinů byla změřena specifická a relativní aktivita se substráty uridinem a 5-methyluridinem a pomocí nelineární regrese podle Michaelise a Mentenové a dvojitě reciprokého vynesení podle Lineweavera a Burka byly ze saturačních křivek stanoveny Km, Vlim a relativní poměr Vlim/Km.The aim of this Master thesis was to characterize the function of aldehyde dehydrogenases from family 2 in Hordeum vulgare and nucleosidases from families 2 and 3 in Zea mays. Within the theoretical part of this work, the literature restrictions dealing with the issue of aldehyde dehydrogenases, their occurrence and function in plants and animals. Another was described the metabolism of purine and pyrimidine bases and the functions of nucleosides in plants. The experimental part includes a search for ALDH genes in the barley genome in the web databases GenBank, Phytozome and UniProt using BLAST software, where 15 genes were found. This part was also focused on cloning, expression and purification of ALDH2 protein from barley. The optimal pH for this enzyme was determined and the substrate specificity was measured. Based on the results, six substrates were selected, for which saturation curves were determined and from them the kinetic parameters Km, Vlim and the relative ratio Vlim/Km were calculated. Finally, the enzymes NRH2b and NRH3 from maize were expressed and purified. The specific and relative activity of these proteins was measured with the substrate uridine and 5-methyluridine and determined by Km, Vlim and relative ratio Vlim / Km from saturation curves by non linear regression according to Michaelis and Menten and double reciprocal yield according to Lineweaver and Burke.
