Diplomová práce se v teoretické části zabývá tématem fotoreceptorů a jejich zapojení ve fotomorfogenezi rostlin, aniontových kanálů a jejich inhibitorů. Cílem experimentální části bylo metodou kvantitativní PCR (qPCR) vyšetřit expresi genů CLCb a CLCc, kódujících chloridové kanály, v hypokotylech mutanta rajčete 7B-1 a kontrolního genotypu cv. Rutgers v závislosti na světelných podmínkách (tma a modré světlo) a přítomnosti inhibitorů aniontových kanálů NIF a DIDS. Provedené experimenty ukázaly, že u obou genotypů modré světlo i přidaný inhibitor NIF mají inhibiční účinek na expresi genu CLCb. Exprese genu CLCc je rovněž snížena vlivem modrého světla, ovšem přidáním inhibitoru NIF je inhibiční vliv modrého světla na expresi eliminován. Je tedy pravděpodobné, že NIF-sensitivní aniontové kanály jsou nějakým způsobem zapojeny v signální dráze modrého světla regulující růst hypokotylu rajčete. V experimentech s inhibitorem DIDS byly získány nereprodukovatelné výsledky a není proto možné učinit jednoznačné závěry o zapojení DIDS-sensitivních kanálů ve fotomorfogenezi rostlin rajčete.The diploma thesis consists of two parts. The first part includes theoretical background for the problems of photoreceptors and their role in plant photomorphogenesis, of anion channels and their inhibitors. The aim of the experimental part was to study, using quantitative PCR (qPCR), expression of genes CLCb and CLCc coding for chloride channels in hypocotyl of the tomato mutant 7B-1 and the control genotype cv. Rutgers as a function of light conditions (darkness and blue light) and in the presence of anion channel inhibitor NIF or DIDS. The results show that blue light and NIF have inhibitory effect on CLCb expression. The expression of the CLCc gene is lowered by blue light as well, but adding of NIF abolishes the effect of blue light on expression of the CLCc. It is therefore likely that NIF-sensitive anion channels are somehow involved in blue light signaling regulating growth of tomato hypocotyls. In the experiments with DIDS, the results were not reproducible and therefore it is not possible to make conclusions about the role of DIDS-sensitive channels in tomato plant photomorphogenesis.
