Cieľom tejto bakalárksej práce bolo nájdenie polymorfných mikrosatelitov u pelikána kučeravého (Pelecanus crispus) pomocou cross-species PCR amplifikácie. V rámci teoretickej časti bolo popísané zaradenie pelikána kučeravého do systému, bol popísaný rad veslonožče, čeľaď pelikánovité a nakoniec samotný pelikán kučeravý. Ďalšia časť bola zameraná na mikrosatelity, ich distribúciu v rôznych genómoch, mutácie a použitie najmä pri prevedení cross-species PCR amplifikácie. V poslednej časti boli opísané polymorfné mikrosatelity pre konkrétne druhy z radu rúrkonoce. Praktická časť zahŕňala testovanie 213 mikrosatelitov na DNA 6 nepríbuzných jedincoch pelikána kučeravého, pričom 207 bolo izolovaných de novo pre druhy patriace do radu rúrkonosce a zvyšných 6 mikrosatelitov, vykazovalo polymorfizmus pre tieto druhy, hoci boli izolované od zástupcov patriacich do radov bahniaky a spevavce. Pomocou cross-species PCR amplifikácie bolo nájdených 12 polymorfných mikrosatelitov, pri čom jeden z nich obsahoval dve miesta polymorfizmu. Jeden mikrosatelit neamplifikoval PCR produkt. Zvyšných 200 mikrosatelitov bolo monomorfných.The aim of this bachelor thesis was finding polymorphic microsatellites in Dalmatian pelican (Pelecanus crispus) using cross-species PCR amplification. Within the theoretical part, was described systematic classification of the Dalmatian pelican, the Pelecaniformes, the family Pelecanidae and finally the Dalmatian pelican. Next part was focused on microsatellites, their distribution in different genomes, mutations and application, especially, with using cross-species PCR amplification. In the last part, polymorphic microsatellites and their isolation for specific species from the order of Procellariiformes were described. The practical part included testing of 213 microsatellites on DNA of 6 unrelated individuals of Dalmatian pelican, whereas 207 was isolated de novo for species belonging to the order Procellariiformes a remaining 6 microsatellites were showed to be polymorphic for these species, altough, they were isolated from individuals from the orders Charadriiformes and Paseriiformes. Using cross-species PCR amplification, 12 polymorphic microsatellites were found, while the one of them had two regions of polymorphism. One microsatellite did not amplified PCR product. Remaining 200 microsatellites were monomorphic.
