Byl studován cytochrom c (cyt c) izolovaný z koňského a telecího srdce. Z obou vzorků byly připraveny apoformy, jenž byly následně podrobeny spolu s nativními cyt c elektrochemickým analýzám. Také byla studována jejich stabilita pomocí elektroforézy a Westernova přenosu. Eliminace hemu v případě apoforem byla potvrzena LC-MS analýzou. Elektroforéza na polyakrylamidovém gelu v přítomnosti denaturačního činidla prokázala, že nedochází k degradaci proteinu během izolace. Dále bylo prokázáno, že monoklonální (Ab A-8) a polyklonální (Ab H-104) protilátky se váží jak na nativní cyt c, tak i na strukturu apocytochromů. Při studiu redoxních vlastností byl identifikován redukční signál hemu (pík HM) i samotného proteinu (pík H), čímž bylo možné od sebe rozpoznat nativní cyt c od apocytochromů. Pík H apocytochromů byl podstatně vyšší než u nativních cyt c. Příčinou je denaturace apocytochromu a s tím spojené rozvolnění struktury, jenž bylo potvrzeno pomocí nativní elektroforézy na polyakrylamidovém gelu. Také byly pozorovány oxidační signály cyt c a apoforem při použití voltametrických metod. Předložené výsledky přispívají k pochopení redoxních vlastností konjugovaných proteinů. Mohou sloužit jako základ pro studium jejich struktury pomocí elektrochemických senzorů.Cytochromes c (cyt c) and their apoforms from bovine and equine heart were studied. Both cyt c native forms and apoforms were isolated and subjected to electrochemical analysis at carbon and Hg-containing electrodes. For the apoforms, the elimination of heme was confirmed by LC-MS and inevitable degradation of the samples during isolation was investigated by denaturing polyacrylamide gel electrophoresis together with protein identification by monoclonal (Ab A-8) and polyclonal (Ab H-104) antibodies. Using chronopotentiometry the reduction signal of heme (peak HM) and the signal of the protein moiety (peak H) were observed. The peak H of the apocytochrome forms was significantly higher than that of the native cyt c forms. The reason for this is that the apoforms of cyt c undergo a relaxation of their structure compared to the native proteins, which was confirmed by native polyacrylamide gel electrophoresis. In addition to this, both structural forms (native cyt c vs. apocyt c) also gave a different electrooxidation pattern using voltammetry. The results presented here could contribute to understanding the redox properties of conjugated proteins in general. They could also serve as a starting point for further studies on the structure and function of cytochromes using electrochemical sensors.
