Telomery jsou specifické nukleoproteinové struktury na koncích eukaryontních chromozomů. Zabezpečují stabilitu konců lineárních chromozomů tím, že řeší koncový replikační problém a maskují konce chromozomů před buněčnými mechanismy opravy dvouvláknových zlomů DNA. Klíčovou roli v údržbě telomer hraje enzym telomeráza. Tento enzym syntetizuje telomerickou DNA sestávající z krátkých tandemových repetic a ovlivňuje tak jejich délku. Dostatečná délka telomer je základním předpokladem jejich správné funkce, přičemž zkrácení pod kritickou délku způsobí buněčnou senescenci a smrt buňky. V posledních letech byly ovšem objeveny i takzvané netelomerické funkce tohoto enzymu, které ovlivňují regulaci buněčné smrti, mohou regulovat transkripci a hrají i roli při odpovědi na poškození DNA. Tyto alternativní funkce by mohly být v budoucnu využity při genové terapii a mít důležitou roli při léčbě rakoviny. Jednou z těchto alternativních funkcí je zapojení katalytické podjednotky telomerázy TERT do signální dráhy Wnt, která může za přítomnosti specifického proteinu beta-kateninu regulovat genovou transkripci a zpětnou vazbou regulovat expresi TERT. Součástí experimentální části je pozorování sensitivity telomerizovaných buněčných linií na specifické inhibitory enzymů z rodiny poly (ADP-ribóza) polymeráz (PARP), zejména na inhibitor KU- 58948, který působí jako inhibitor PARP1. Cílem bylo prozkoumat podíl Wnt dráhy indukované ektopickou expresí TERT na rezistenci vůči inhibitorům PARP.Telomeres are specific nucleoprotein structures at the ends of eukaryotic chromosomes. They maintain the stability of the ends of linear chromosomes by providing solution to the end replication problem and masking the ends of chromosomes from the cellular DNA repair mechanisms. A key role in telomere maintenance is played by the enzyme telomerase. Telomerase synthesizes telomeric DNA consisting of short tandem repeats and thus affects their length. Sufficient telomere length is an essential prerequisite for their proper function while shortening below a critical length causes cellular senescence and cell death. Interestingly, novel non - telomeric function of the telomerase enzyme have been discovered - which affect the regulation of cell death, transcription of specific genes and DNA damage response. These alternative functions could be used in the future for gene therapy and treatment of cancer. One of the newly discovered alternative functions is the involvement of telomerase catalytic subunit TERT in the Wnt signaling pathway. Along with the key regulator of the pathway beta-catenin TERT regulates transcription of Wnt genes and creates feedback look that in turn regulates TERT expression via the Wnt pathway. The aim of the experimental part of the work is to test the sensitivity of certain cell lines ectopically expressing telomerase to the specific inhibitor of the poly (ADP ribose) polymerase (PARP1) enzyme. In particular, we wanted to assess the role of TERT induced Wnt signalling in resistence to PARP inhibitors.
