Cyklin dependentní kinasa 4 (CDK4) patří do skupiny Ser/Thr proteinkinas esenciálních pro regulaci buněčného cyklu. Za fyziologických podmínek aktivní komplex CDK4/cyklin D reguluje progresi G1 fáze a různé deregulace genu pro CDK4 nebo souvisejících proteinů jsou spojovány s tumorgenezí. V současné době je CDK4 považována za slibný terapeutický cíl, dokonce již bylo schváleno několik ATP-kompetitivních inhibitorů pro léčbu metastatického karcinomu prsu v kombinaci s endokrinní terapií. Praktická část je věnována studiu účinku nových CDK4 inhibitorů a jejich vlivu na životaschopnost linií karcinomu prsu v kombinaci s edokrinní a anti-HER2 terapií. Pro vypracování praktické části byly použity metody: průtoková cytometrie, western blotting, CETSA, -galaktosidasový test a stanovení cytotoxicity resazurinem. Testované inhibitory vykazovaly na buněčné úrovni zastavení buněčného cyklu v G1 fázi spojené s poklesem hladiny fosforylovaných forem proteinu RB a indukovaly navození senescentního fenotypu nádorových buněk, obdobně jako inhibitor palbociclib. V kombinačních experimentech látky vykazovaly spíše aditivní účinky s anti-HER2 a endokrinní terapií. Testované inhibitory prokázaly na buněčných liniích odlišný profil cytotoxicity, což jen vyzdvihuje význam charakterizace nespecifických účinků látek, které mohou mít dále dopad na klinické úrovni. Práce poukazuje také na rizika vzniku lékové rezistence a shrnuje nástroje zamezení vzniku prostřednictvím kombinační terapie.Cyclin-dependent kinase 4 (CDK4) belongs to the group of Ser/Thr protein kinases essential for cell cycle regulation. Under physiological conditions, the active CDK4/ cyclin D complex regulates G1 phase progression and various CDK4 gene deregulation or related proteins are associated with tumorigenesis. CDK4 is currently considered a promising therapeutic target, and several ATP-competitive inhibitors have even been approved for the treatment of metastatic breast cancer in combination with endocrine therapy. The practical part is devoted to the study of the effect of new CDK4 inhibitors and their effect on the viability of breast cancer lines in combination with endocrine and anti-HER2 therapy. The following methods were used to develop the practical part: flow cytometry, western blotting, CETSA, -galactosidase assay and determination of cytotoxicity by resazurin. At the cellular level, the inhibitors tested showed G1 phase cell cycle arrest associated with decreased levels of phosphorylated forms of the RB protein and induced the induction of a senescent phenotype of tumor cells, similar to the palbociclib inhibitor. In combination experiments, the substances showed an additive effects with anti-HER2 and endocrine therapy. The tested inhibitors showed a different cytotoxicity profile on the cell lines, which only highlights the importance of characterizing the non-specific effects of the substances, which may further have an impact at the clinical level. The work also points out the risks of drug resistence and summarizes the tools to prevent the development through combination therapy.
