Kvartérní benzo[c]fenantridinové alkaloidy jsou látky vykazující širokou biologickou a fyziologickou aktivitu. Tyto látky se využívají jako biopesticidy, v boji proti paradentóze, mají nemalý význam při apoptotických procesech, v léčbě rakovinných buněk, ovlivňují řadu enzymů a mají mnoho dalšího využití. Při interakci těchto látek s DNA může docházet ke kovalentní vazbě nebo nekovalentním interakcím, kde rozlišujeme mezi interkalací nebo vazbou do žlábku. Pro charakterizaci strukturních analogů chelerytrinu a sanguinarinu, tedy NK-109 a redukované formy NK-109, byly využity metody fluorescenční spektroskopie a spektrofotometrie. Byla proměřena fluorescenční a absorpční spektra pro různé koncentrace DNA a různé koncentrace zkoumaných alkaloidů, na základě kterých byly zjištěny cenné informace ? rozpustnost v daných rozpouštědlech, závislost fluorescence alkaloidu na přítomnosti metanolu a stechiometrie vazby na DNA.Quaternary benzo[c]phenanthridine alkaloids are compounds, which exhibit a wide range of biological and physiological activities. These species are used as biopesticides, against periodontisis, they have a significant role in apoptotic processes, in the treatment of cancer cells, they affect a variety of enzymes and have many other applications. The interaction of these compounds with DNA may be covalent bond or non-covalent interaction, where we distinguish intercalation process and binding into the groove. For characterization of the chelerythrine and sanguinarine analogs termed NK-109 and reduced form of NK-109, we utilized the fluorescence spectroscopy and spectrophotometry. We analyzed fluorescence and absorption spectra for different concentrations of DNA and various concentrations of alkaloids. This study revealed valuable information about a solubility in some solvents, dependence of fluorescence on methanol presence and DNA-binding stoichiometry.
