Medicago sativa patří mezi luštěniny, které dokážou interagovat a vytvářet symbiotický vztah s půdními bakteriemi rodu Rhizobium. Tento vztah spočívá ve vytváření specializovaných orgánů (tzv. hlízek) na kořenech rostlin, v nichž bakterie redukují atmosférický dusík na amoniak. Pro zahájení interakce mezi rostlinou a bakterií jsou klíčové mitogenem-aktivované proteinkinasy (MAPK), podílející se na přenosu signálu z vnějšího prostředí. U M. sativa byla v rámci popsaných MAPK identifikována SIMK (stresem indukována MAPK), která dokáže ovlivňovat stav a dynamiku aktinového cytoskeletu a je aktivní zejména během vytváření a vývoje kořenových vlásků, což může ovlivnit také efektivitu procesu tvorby hlízek. Cílem této bakalářské práce byla regenerace kontrolních rostlin M. sativa Regen-SY a transgenních rostlin M. sativa SIMK RNAi v in vitro podmínkách procesem somatické embryogeneze. Dále fenotypická charakterizace a srovnání kontrolních a transgenních rostlin M. sativa prostřednictvím měření délky kořenové soustavy a počítání laterálních kořenů. Po aplikaci půdních bakterií Sinorhizobium meliloti na kořeny rostlin byla vyhodnocována efektivita tvorby hlízek.Medicago sativa is one of the legumes that can interact and form a symbiotic relationship with soil bacteria of the genus Rhizobium. This relationship includes the formation of specialized organs (so-called nodules) on the roots of plants, in which bacteria reduce atmospheric nitrogen to ammonia. Mitogen-activated protein kinases (MAPKs), involved in signal transduction from the environment, are key components of plant-bacterial interactions. In M. sativa, SIMK (stress-induced MAPK) has been identified within the described alfalfa MAPKs, which can influence the state and dynamics of the actin cytoskeleton. SIMK is active especially during the formation and development of root hairs, which may also affect the efficiency of the nodule formation process. The aim of this bachelor thesis was the regeneration of control Regen-SY and transgenic SIMK RNAi alfalfa plants in vitro by the process of somatic embryogenesis. Furthermore, phenotypic characterization and comparison of control and transgenic M. sativa plants was performed by measuring the length of the root system and counting the lateral roots. After application of soil bacteria Sinorhizobium meliloti to plant roots, the effectiveness of nodulation was evaluated.
