Aryl uhľovodíkový receptor (AhR) je ligandom aktivovaný transkripčný faktor, ktorý zohráva kľúčovú úlohu v celej škále fyziologických procesov. Jedným z široko využívaných nástrojov pri štúdiu transkripčnej aktivity AhR sú luciferázové reportérové systémy. V predkladanej diplomovej práci bola charakterizovaná nová reportérová bunková línia LS174T-AhR-luc na stanovovanie transkripčnej aktivity AhR. Na základe experimentov bolo zistené, že po aplikácii modelových ligandov AhR došlo k indukcii luciferázovej aktivity, ktorej intenzita bola závislá od koncentrácie ligandov. Bunky LS174T-AhR-luc si zachovali schopnosť inducibility luciferázovej aktivity ako po kryoprezervácii a uchovávaní pri nízkej teplote, tak aj po dlhodobom udržovaní v kultivačnom médiu. Rovnako došlo aj k zachovaniu morfológie buniek v porovnaní s rodičovskou líniou LS174T. Testy využívajúce inhibítor AhR CH-223191 potvrdili, že indukcia luciferázovej aktivity prebiehala špecificky, v dôsledku pôsobenia AhR ligandu. Takisto, aj testovanie vplyvu ligandov vybraných jadrových a steroidných receptorov odhalilo, že indukcia luciferázovej aktivity bola selektívna voči ligandu AhR. Získané výsledky tak ukázali, že nová reportérová línia LS174T-AhR-luc spĺňa všetky požadované kritériá pre jej úspešné využívanie v experimentálnej praxi.The aryl hydrocabon receptor (AhR) is a ligand-activated transcription factor that plays a key role in various physiological processes. One of the widely used tools for studying AhR transcriptional activity are luciferase reporter systems. In the present diploma thesis, a novel reporter cell line for the assessment of AhR transcriptional activity, LS174T-AhR-luc, was characterized. Based on the experiments, it was found that the application of AhR model ligands resulted in the dose-dependent induction of luciferase activity. LS174T-AhR-luc cells retained the inducibility of luciferase activity both, after the cryopreservation and storage at low temperature, and after the long-term maintenance in the culture medium. Morphology of the cells was also preserved compared to parental cell line LS174T. Assays using the AhR inhibitor CH-223191 confirmed that the induction of luciferase activity was a specific result of AhR ligand action. Also, testing the effects of ligands of selected nuclear and steroid receptors revealed that the induction of luciferase activity was selective for the AhR ligand. The obtained data showed that the novel reporter cell line LS174T-AhR-luc meets all the required criteria for its successful use in experimental practice.
