Auxin je klíčovým rostlinným hormonem, který spolu s dalšími fytohormony řídí správný růst a vývoj rostlin. Přesná časová a místní regulace auxinové distribuce v rámci jednotlivých rostlinných pletiv v kombinaci s percepcí auxinového signálu na buněčné úrovni stojí za širokým spektrem biologických účinků auxinů. Tato práce shrnuje charakterizaci nových syntetických strukturních derivátů auxinů, které mohou sloužit jako užitečný nástroj pro studium molekulárních mechanismů působení auxinů. Auxinové fluorescenční deriváty mohou pomoci odhalit principy distribuce a transportu auxinů v rámci jednotlivých vývojových procesů. Látky vykazující funkci selektivních agonistů různých auxinových signálních drah mohou pomoci objasnit roli jednotlivých ko-receptorových systémů v rámci auxinem vyvolaných fyziologických odpovědí. Dále byly vyvinuty citlivé purifikační a detekční metody využívající kapalinovou chromatografii ve spojení s hmotnostní detekcí za účelem studia metabolismu syntetických derivátů v rostlinách. Závěrem, nová auxinová analoga, společně s vyvinutými technikami ke studiu jejich stability v živém systému, představují inovativní přístup studia auxinové fyziologie, který pomůže odhalit mechanismy stojící za komplexním souborem biologických účinků auxinů ve vývoji rostlin.Auxin is a crucial hormonal player that in cooperation with other phytohormones regulates proper plant growth and development. Its precise spatiotemporal distribution in specific tissues together with auxin perception at a cellular level underlies a plethora of auxin actions. This thesis presents a characterization of new synthetic structural auxin analogues that can serve as a chemical tool for investigation of the molecular mechanisms of auxin physiology. The fluorescent auxin derivatives can help to determine auxin transport sites within various developmental processes while the selective auxin agonists can elucidate the roles of specific auxin co-receptors in auxin-regulated responses. Sensitive purification and detection methods based on liquid chromatography-mass spectrometry (LC-MS) technique have also been developed to evaluate the metabolization of synthetic auxin analogues in planta. Taken together, the novel synthetic auxin derivatives together with MS-based methods for evaluation of their stability in vivo represent a powerful tool for unravelling the complexity of auxin actions.
