Cílem této bakalářské práce bylo zjištění přesných pozic zlomů DNA, vedoucích k velkým intragenovým přestavbám v genech reparačního systému BRCA1, BRCA2 a CHEK2. Práce vychází z intragenových přestaveb, které byly zachyceny v diagnostické laboratoři při rutinních vyšetřeních a je rozdělena do dvou částí. Teoretická část se věnuje literární rešerši jednotlivých genů, jejich struktuře a funkci a shrnuje záchyty různých přestaveb v těchto genech v rámci Evropy i ve světě. Zároveň uvádí možné důvody vzniku těchto změn v DNA ve vztahu k přítomnosti repetitivních sekvencí v genech. Je zde také stručně shrnutá struktura lidského genomu, zvláště pseudogenů a Alu elementů, které se v těchto genech vyskytují. V poslední části teoretického úvodu jsou charakterizovány metody molekulární biologie využívané při analýze intragenových přestaveb. V experimentální části jsou uvedeny výsledky charakterizace zlomových míst ve výše zmíněných třech tumor supresorových genech, včetně obrázků elektroforetických gelů, Sangerových sekvenací a popisu oblastí tvořících přestavbu. V rámci práce byla popsána také intragenová delece genu BRCA2, která byla zachycena v ČR poprvé v laboratoři v Novém Jičíně. Ze čtyř záchytů zjištěných pomocí metody multiplex ligation-dependent probe amplification jsme u tří případů metodami molekulární biologie přesně popsali zlomová místa.The point of this bachelor thesis was to find exact positions of DNA breakpoints, which leads to large genomic rearrangement (LRG) in DNA repair genes BRCA1, BRCA2 and CHEK2. This thesis is based on LRGs, which were detected in the diagnostic laboratory during routine examinations and is divided into two parts. The theoretical part is focused on the literary search of the above mentioned genes, their structure and function, and summarizes the results of various LRGs detected in these genes within Europe and around the world. Thesis subsequently states the possible reasons for these changes in DNA in relation to the presence of repetitive sequences in the genes and briefly summarizes the structure of the human genome, especially pseudogens and Alu elements found in these genes. In the last part of the theoretical introduction, the molecular biology methods used in the analysis of intragenic reconstructions are characterized. In the experimental part, the results of the characterization of the breakpoints in three tumor suppressor genes, including the images of the electrophoretic gels, the Sanger sequencing results and the description of the regions forming the rearrangement, are presented. The work also described the intragenic deletion of the BRCA2 gene, which was detected for the first time in Czech Republic. Of the four LRGs detected using the multiplex ligation-dependent probe amplification method, in three cases, molecular biology methods accurately described breakpoints.
