Súčasný tlak súvisiaci s nárastom bakteriálnej rezistencie voči antibiotikám vedie výskum k hľadaniu nových možností liečby bakteriálnych ochorení. Ako jednou z možností riešenia tohto problému sa javí využitie antimikrobiálnych peptidov, ktoré sú známe vďaka svojmu širokospektrálnemu pôsobeniu voči celej rade bakteriálnych kmeňov. Navyše, metódy rastlinnej biotechnológie s využitím obilnín, ako expresných systémov, umožňujú produkciu antimikrobiálnych peptidov vo veľkom meradle a s možnosťou jeho dlhodobého uskladnenia v biologicky aktívnej forme v inertnom prostredí endospermu obilných zŕn. V experimentálnej časti diplomovej práce bola analyzovaná T1 a T2 generácia transgénnych rastlín jačmeňa siateho (Hordeum vulgare L., cv. Golden Promise) exprimujúcich rekombinantný antimikrobiálny peptid LL-37 zo skupiny katelicidínov. U transgénnych rastlín bol sledovaný vplyv expresie rekombinantného peptidu na fenotyp. Prítomnosť transgénu v genóme rastlín T2 generácie bol potvrdený pomocou metódy konvenčnej PCR. Následne bol rekombinantný peptid detegovaný vo vegetatívnych orgánoch a v zrnách transgénnych rastlín T1 a T2 generácie na proteínovej úrovni pomocou metódy Western blot. Zároveň bolo overené uskladnenie rekombinantného peptidu LL 37 v rámci endospermu zrelých zŕn metódou imunolokalizácie. Taktiež bola overená biologická aktivita rekombinanntého katelicidínu voči baktériam Escherichia coli TOP10 a Micrococcus luteus. V neposlednom rade bola vykonaná dvojkroková purifikácia rekombinantného produktu extahovaného zo zŕn transgénnych rastlín jačmeňa založenej na metalo-chelátovej afinitnej chromatografii.The current pressure associated with the increase of bacterial resistance against antibiotics leads to research to find new ways of treating bacterial diseases. One of the possibilities of solving this problem is the use of antimicrobial peptides well known for their broad-spectrum activity against a various bacterial strain. In addition, methods of plant biotechnology using cereals, as expression systems, allow the production of antimicrobial peptides in large scale. Also, targeting and production of antimicrobial peptides in the inert environment of the grain endosperm allows its long-term storage in a biologically active form. In experimental part of this diploma thesis, T1 and T2 generations of transgenic barley plants (Hordeum vulgare L., cv. Golden Promise) expressing recombinant antimicrobial peptide LL-37 from the group of cathelicidins were analysed. At first, effect of recombinant product expression on phenotype of transgenic barley was studied. Presence of transgene in plant's genome of T2 plants was confirmed by conventional PCR. Subsequently, the recombinant peptide was detected in the vegetative organs and in the grains of the T1 and T2 transgenic plants at the protein level using the Western blot method. Furthermore, the storage of the recombinant peptide LL-37 within the mature grains endosperm was confirmed by the immunolocalization. The biological activity of recombinant peptide against Escherichia coli TOP10 and Micrococcus luteus was also verified. Finally, two step purification of the recombinant product extracted from transgenic barley grains was purified by two step purification process based on metal-affinity chromatography.
