Hlístice patří mezi nejpočetnější mnohobuněčné organismy, které obývají široké spektrum biotopů. Vyznačují se řadou různých životních strategií, existují jako parazité nebo volně žijící druhy. Přesto, že jich dodnes bylo popsáno asi 30 000 druhů, jejich výzkum není ani zdaleka u konce. Zaměřením této bakalářské práce byla molekulární identifikace vybraných neznámých druhů hlístic, které již byly morfologicky zařazeny do určitých rodů. K tomuto účelu byly amplifikovány fragmenty DNA hlístic jaderného úseku LSU ("large subunit") a mitochondriálního genu COI. Tyto fragmenty byly zaklonovány do plasmidu a sekvence fragmentů byla komerčně přečtena. Pro získané sekvence byly pomocí programu BLAST nalezeny homologní sekvence, které se v současné době nachází v nukleotidové databázi NCBI. Šest z osmi získaných sekvencí odpovídalo sekvencím hlístic, jedna z nich se s nalezenou plně shodovala. Zbývající dvě sekvence byly identifikovány jako houby, což naznačuje možnost kontaminace.Nematodes belong to the one of the largest group of multicellular organisms that occupies a wide range of biotopes. They developed a broad variety of life strategies, as for example parasitic or free living life style. There has been described about 30 000 species till now, however, this number is far from complete. This bachelor thesis has been focused on molecular identification of unknown nematode species, which have already been morphologically classified into certain genus. For this purpose the DNA fragments of nuclear region LSU (large subunit) and mitochondrial gene COI were amplified. These fragments were inserted into the plasmid and the sequences of fragments were commercially sequenced. The sequences that have been acquired were used to search for the homologous sequences in the nucleotide database of NCBI using the program BLAST. Six of eight sequences were homologous to the sequences of nematodes found in database, one of them was found to be identical. The remaining two sequences were identified as fungi, suggesting the possibility of contamination.
