Adenosindeaminasy patří do amidohydrolasové rodiny (EC 3.5.4.-) a katalyzují ireverzibilní deaminaci adenosinu a 2-deoxyadenosinu. Byly identifikovány v celé škále eukaryot i prokaryot. V odborné literatuře nejsou doposud uvedeny žádné informace popisující přítomnost a aktivitu těchto enzymů u rostlin, a to i přesto, že v genomových databázích existují geny kódující předpokládané rostlinné adenosindeaminasy. Tyto enzymy by mohly hrát klíčovou roli při vývoji rostlin. Prezentovaná diplomová práce navazuje na již ukončenou bakalářskou práci se zaměřením na potvrzení aktivity předpokládané rostlinné adenosindeaminasy. V první řadě byla tato práce zaměřena na nalezení vhodných purifikačních podmínek pro SUMO modifikovanou adenosindeaminasu s následným kvantitativním odštěpením His-SUMO kotvy. Následně byla měřena aktivita třemi principově odlišnými metodami, z nichž jako nejvhodnější byla nakonec vybrána UHPLC. Aktivita adenosindeaminasy byla porovnávána s lidskou adenosindeaminasou a připravenou adenosindeaminasou podle Pospíšilová et al., 2008. U předpokládané rostlinné adenosindeaminasy byly finálně stanoveny kinetické parametry vůči pravděpodobně hlavnímu substrátu AMP.Adenosine deaminases belong to the amidohydrolase family (EC 3.5.4.-) and catalyse the irreversible deamination of adenosine and 2'-deoxyadenosine. They have been identified in a wide range of eukaryotes and prokaryotes. In the genome databases have been identified many genes, which are annotated as putative plant adenosine deaminases. But until now the presence and activity of these enzymes in plants was not confirmed. It is possible that they can play a crucial role in plant development. The presented diploma thesis continues with work, which was done during finished bachelor thesis. The main ambition of the thesis was to confirm the putative plant adenosine deaminase activity. Primarily, this work was focused on finding the best purification conditions for SUMO-modified adenosine deaminase what was followed by quantitative cleavage of His-SUMO tag from the enzyme. Subsequently, the activity was measured by three different methods. As the most suitable was selected UHPLC. Adenosine deaminase activity was measured and compared with activity of human adenosine deaminase and adenosine deaminase prepared according to protocol describe in work Pospíšilová et al., 2008. Finally the kinetic parameters against probably the main substrate AMP have been determined.
