Studium interakce rostlin z čeledi Cucurbitaceae (tykvovité) a jejich patogenu, oomycety Pseudoperonospora cubensis, způsobující plíseň okurky, je z hlediska velkého ekonomického významu tykvovitých plodin a rozšíření P. cubensis dlouhodobě velmi důležité a potřebné. Zaměřuje se mj. na identifikaci nových efektivních zdrojů rezistence (mezi pěstovanými i planými zástupci čeledi Cucurbitaceae), využitelných ve šlechtění tykvovitých na rezistenci vůči P. cubensis. Analýza dat testování rezistence 97 genotypů planých a plevelných taxonů Cucurbita spp. vůči 11 izolátům P. cubensis (2001) potvrdila možnost existence genů rezistence v rámci rodu Cucurbita. Výsledné reakce testovaného souboru mají rasově specifický charakter. V návaznosti na toto testování byl proveden screening 22 genotypů Cucumis melo na odolnost vůči 8 izolátům P. cubensis. Díky převaze náchylných reakcí vzorků C. melo nebyly objeveny nové zdroje rezistence, rasová specifičnost interakce C. melo a P. cubensis však byla evidentní. Výsledky této diplomové práce poskytují nové informace o vztahu hostitele a patogenu, které jsou použitelné v dalším výzkumu.From the viewpoint of great economic significance of cucurbit crops and distribution of their pathogen, oomycete Pseudoperonospora cubensis, research of interaction between plants of the Cucurbitaceae and P. cubensis, which causes cucurbit downy mildew, has been very important and required for a long time. The research has been also focused on identification of new efficient sources of resistance (among cultivated and wild Cucurbitaceae plants), that could be exploited in breeding of cucurbits for resistance against P. cubensis. Analysis of screening data of 97 genotypes of wild and weedy Cucurbita spp. for resistance against 11 isolates of P. cubensis (2001) supported the expectation of existence of resistance genes in the genus Cucurbita. Final reactions of studied set were considered as race-specific. Subsequently, 22 Cucumis melo genotypes were screened for resistance against 8 isolates of P. cubensis. Because of dominance of susceptible reactions of C. melo accessions, new sources of resistance wasn't found, but the race specificity of C. melo - P. cubensis interaction was evident. The results of this master thesis provide new pieces of knowledge about host-pathogen relationship that are useful for further research.
