Nesyt indomalajský (Mycteria leucocephala) společně se třemi dalšími druhy (nesyt lesní, nesyt africký a nesyt bílý) tvoří trib Mycteriini spadající pod rod nesytů, čeled čápovitých a řád brodivých. Tento elegantní pták s pestobarevným peřím byl kdysi hojně se vyskytující. Avšak dnes je veden jako téměř ohrožený druh. Z tohoto druhu doposud nebyly izolovány žádné mikrosatelity. Teoretická část mé bakalářské práce pojednává o nesytu indomalajském. Věnovala jsem se systematickému zařazení tohoto druhu, dále pak způsobu jeho života, lovu, hnízdění a péči o mládě a také místům jeho výskytu. Dále jsem shromáždila informace o mikrosatelitech, které jsem na tomto druhu testovala. Psala jsem o jejich původu a charakteristikách. V praktické části jsem za pomocí cross ? species PCR amplifikace testovala všechny dosud publikované primery pocházející z řádu brodiví a všechny doposud nalezené primery z řádu potápek a potáplic. Dále jsem pak otestovala vybrané primery od druhů z řádu dlouhokřídlých, vrubozobých, tučňáků a sudokopytníků. Otestovala jsem tak celkem 199 párů primerů, z nichž bylo 21 polymorfních, dají se tak využít pro další studium nesyta indomalajského.The Painted Stork (Mycteria leucocephala) together with three other species (the Milky Stork, the Yellow-billed Stork and the Wood Stork) form a trib Mycteriini, which belongs under the Mycteria genus, the Ciconiidae family and the Ciconiiformes order. This elegant bird with colourful feathers was once abundant. But today it is listed as near threatened species. For this species have so far been isolated no microsatellites. The theoretical part of my bachelor thesis discusses about the Painted Stork. I wrote about a systematic classification of this species, then about a way of life, hunting, breeding and taking care of the brood and also I mentioned about it?s habitat. I also gathered information about microsatellites, which I tested on this species. I wrote about their origin and characteristics. In the practical part I used cross ? species PCR amplification to test all previously found primers derived from the order of Ciconiiformes and all previously found microsatelites from Podicipediformes and Gaviiformes order. Then I tested selected primers derived from the Charadriiformes, Sphenisciformes, Anseriformes and Artiodactyla orders. I have tested 199 pairs of primers. 21 of them were polymorphic, and they can be used for further study of the Painted Stork.
