Tato bakalářská práce je zaměřena na studování vlivu obsahu kluzných látek při tabletování farmaceutických směsí. V této práci je kluznou látkou stearan hořečnatý v různých koncentracích a léčivou látkou kyselina listová. V průběhu práce byl studován vliv obsahu kluzné látky a lisovací síly na disoluční profily účinné látky, pevnost, oděr, hmotnost, výšku a rozpad tablet. Experimentálně bylo zjištěno, že se zvyšujícím se obsahem stearanu hořečnatého dochází ke snížení pevnosti. Dále bylo dokázáno, že závislost výšky tablet na lisovací síle projevilo opačnou závislost s porovnáním závislosti pevnosti, která je s lisovací silou logicky rostoucí. Klesající závislost dokazuje účinek lisovací síly tabletovacího stroje. Disoluční studie ukázala, že se zvyšujícím obsahem stearanu hořečnatého se disoluce zhoršuje a zpomaluje. Akcelerovaná stabilitní studie po 1 a 3 měsících ukázala, že vzhled a jejich hmotnostní stejnoměrnost byly v normě dle předepsané specifikace. Ani u stanovení obsahu API vyrobených tablet nemá stabilitní studie výrazný vliv. U testování pevnosti tablet se ukázalo, že i po akcelerované době se pevnost tablet zhoršuje s rostoucí koncentrací stearanu hořečnatého.This bachelor thesis is focused on the study of the effect of content of sliding substances during the tabletting of pharmaceutical mixtures. In this thesis, the sliding substance is magnesium stearate in various concentrations and the medicinal substance is folic acid. During the course of the thesis, the effect of content of sliding substance and pressing force on dissolution profiles of active substance, solidity, abrasion, weight, height and decomposition of tablets was studied. It was experimentally found out that with the increasing content of magnesium stearate, the decrease of solidity occurs. It was further proven that the dependence of the height of tablets on pressing force demonstrated the opposite dependence in comparison with the dependence of solidity which logically increases with the pressing force. The decreasing dependence proves the effect of pressing force of the tablet machine. The dissolution study showed that with the increasing content of magnesium stearate, the dissolution worsens and slows down. The accelerated study of stability after 1 and 3 months showed that the apperance and their weight uniformity were within standards according to the prescribed specification. Neither at the determination of content of API of the produced substances, the study of stability does not have any substantial effect. During the testing of solidity of tablets, it was revealed that even after the accelerated period of time the solidity of tablets worsens together with the increasing concentration of magnesium stearate.
