Bakalářská práce je zaměřena na charakterizaci S-nitrosoglutathionreduktasy (GSNOR), klíčového enzymu metabolismu S-nitrosothiolů a formaldehydu, zejména na jeho lokalizaci a fyziologické funkce v živočišných, a především v rostlinných systémech, a úlohu GSNOR v obranné reakci na abiotické i biotické stresové faktory. V úvodních kapitolách teoretické části práce jsou shrnuty poznatky týkající se reaktivních forem dusíku, S-nitrosylace představující mechanismus pro udržování homeostázy S-nitrosothiolů, které slouží jako zásobní nebo transportní formy NO in vivo. V experimentální části práce byla testována připravená primární polyklonální protilátka proti rekombinantnímu proteinu GSNOR z templátu sekvence GSNOR ze Solanum lycopersicum cv. Amateur, který byl izolovaný z E. coli. Bylo stanoveno vhodné ředění protilátky pomocí metody Slot Blot, citlivost a specificita metodou Western blot. Pomocí SDS-PAGE byla stanovena molekulová hmotnost rekombinantního proteinu GSNOR. U vybraných modelových rostlin (tři genotypy Solanum spp., dva genotypy Lactuca sativa, dále u Lactuca serriola, Pisum sativum, Helianthus annuus a dva genotypy Cucumis sativus) byl sledován vliv mechanického poškození listové plochy a teplotního stresu na aktivitu GSNOR. V připravených rostlinných extraktech byla GSNOR lokalizována metodou Western blot.The thesis is focused on the characterization of S-nitrosoglutathione reductase (GSNOR), a key enzyme in the metabolism of S-nitrosothiols and formaldehyde, in particular its localization and physiological function in animal and plant systems, especially in a defensive role GSNOR in response to abiotic and biotic stress factors. In the opening chapters of the theoretical part are summarized the findings of reactive nitrogen species, S-nitrosylation, which represents a mechanism for maintaining homeostasis of S-nitrosothiols, which serve as storage or transport forms of NO in vivo. In the experimental part has been tested primary polyclonal antibody prepared against recombinant protein from the template sequence GSNOR of Solanum lycopersicum cv. Amateur, which was isolated from E. coli. It was determined the appropriate dilution of antibody using the method Slot blot, the sensitivity and specificity by Western blot. By SDS-PAGE was determined molecular weight of recombinant protein GSNOR. In the selected model plants (three genotypes of Solanum spp., two genotypes of Lactuca sativa, Lactuca serriola, Pisum sativum, Helianthus annuus and the two genotypes of Cucumis sativus) was examined the effect of mechanical damage to the leaf surface and heat stress on the activity GSNOR. In the prepared plant extracts GSNOR was localized by Western blot.
