Plíseň okurková je považována za nejzávažnější onemocnění tykvovitých rostlin (Cucurbitaceae), které způsobuje významné hospodářské ztráty na celém světě (Thomas, 1996). Původcem této choroby je obligátní houbový parazit Pseudoperonospora cubensis (Lebeda a Cohen, 2011), který vykazuje rozsáhlý okruh hostitelských rostlin (Lebeda a Urban, 2007; Palti a Cohen, 1980). V posledních letech se hledají cesty jak identifikovat rozdíly jednotlivých kmenů P. cubensis s různými fytopatologickými charakteristikami (např. patotypy), a to i pomocí metod molekulární biologie. V rámci této bakalářské práce byla provedena pomocí metody AFLP detekce variability u souboru 30 izolátů P. cubensis pocházející z České republiky. U každého z testovaných izolátů byl znám jeho patotyp. Genetická analýza izolátů P. cubensis byla provedena pomocí zvolených 13 primerových kombinací. Celkem bylo získáno 387 detekovatelných bandů, na základě kterých byly programem TreeView a FreeTree sestaveny tři dendrogramy. Tyto dendrogramy oddělily izoláty z roku 2009 od ostatních izolátů. Nepodařilo se tedy rozčlenit dané izoláty podle odlišných geografických oblastí. Z těchto výsledků je také patrné, že neexistuje souvislost mezi shlukováním izolátů s podobnými fytopatologickými charakteristikami a jejich AFLP profily. Výsledky této bakalářské práce ukazují na důležitost sledování změn v přírodních populacích P. cubensis, které mohou přispět ve šlechtění rezistentních odrůd tykvovitých plodin, popřípadě k výběru a vývoji fungicidů, které by zmírnily každoroční dopad devastujících epidemií.Downy mildew is considered as the most serious disease of Cucurbitaceae plants which causes the considerable economic losses in the whole world (Thomas, 1996). The etiological agent of this disease is an obligate fungal parasite Pseudoperonospora cubensis (Lebeda and Cohen, 2011) which exhibits an extensive range of host plants (Lebeda and Urban, 2007; Palti and Cohen, 1980). In recent years are searching for ways to identify the differences in individual strains of P. cubensis with different phytopathological characteristics (e.g. the pathotypes) also by using of methods of molecular biology. A detection of the variability was performed within this bachelor thesis by using of method AFLP at the collection of 30 isolates of P. cubensis originated from Czech Republic with its known pathotype characteristics. The genetic analysis of isolates of P. cubensis was performed by using of 13 selected primer combinations. In total 387 AFLP bands were detected and on their basis three dendrograms we constructed using of the TreeView and FreeTree programs. The dendrograms separated the isolates from the 2009 year from the rest of analysed samples. However, segregation of isolates according to different geographical areas has not been confirmed. It is also clear from these results that a clear linkage between the clustering of the isolates with the similar phytopathological characteristics and their AFLP profiles has not been proved. The results of this bachelor thesis demonstrate the importance of change monitoring in natural populations of P. cubensis that can help in breeding of resistant varieties of crops Cucurbitacea or to the selection and the development of fungicides which would reduce the devastating impact of annual epidemics of this pathogen.
