Je známo, že fotosyntéza je proces, který vytvořil kyslíkovou atmosféru, zdrojem kyslíku je fotosyntetické štěpení vody. Užitečná cesta jak prozkoumat mechanismus štěpení vody je měření záblesky indukovného vývoje kyslíku. Krátké a silné záblesky vedou k oscilaci ve vývoji kyslíku s periodicitou čtyřech záblesků. Vhodnou metodou ke zkoumání zmíněné oscilace je kinetické modelování na kterém byl postaven i popis utlumení oscilace vývoje kyslíku a to zavedením dvou kinetických parametrů Besselem Kokem. Význam těchto parametrů nebyl zcela jasný a to zejména kvůli zjednodušeným kinetickým modelům procesu štěpení vody. Tato disertační práce je proto postavena na studiu štěpení vody ve fotosyntéze, zejména na detailnějším matematickém popisu tohoto děje v rámci kinetického modelu. Náš výzkum fotooxidace vody začal analýzou oscilací vývoje kyslíku s periodou čtyř záblesků. Zjistili jsme, že v existujících modelech chybí klíčové spojení mezi procesem štěpením vody a přenosem elektronu skrz fotosystém II (PSII). Jako možné řešení jsme navrhli existenci tzv. přechodných S-stavů, definovaných jako [SnYZox], (n = 0, 1, 2, 3), s rychlostní konstantou jejich vzniku kiSn ~ 1.5 × 106 s-1. Přechodné stavy mají svou regulační a stabilizační roli na celou donorovou stranu PSII a byly definovány jako relaxační mechanismus spojený se změnou rovnováhy P680+YZ ? P680YZox. Oscilace ve vývoji kyslíku jsou tlumené a pro jejich popis byly původně zavedeny parametry ?ztráta? a ?dvojitý přechod?. Za účelem zjištění, co se skrývá za dvojitým přechodem, jsem vytvořil model monomeru a dimeru PSII. Porovnání modelů vedlo k navržení mechanismu kooperace mezi jednotlivými PSII v dimeru jako dílčího zdroje vývoje kyslíku po druhém záblesku. Kooperace se podílí na hodnotě Kokova parametru dvojitý přechod. Konečně, zmíněná kooperace se vyskytuje pravděpodobně jen u sinic, pro které má významný fyziologický význam za nižší intenzity světla. Náš poslední výzkum se týkal redukce vyšších S-stavů ve tmě. Uvažováním všech přenašečů v PSII v jedné stavové proměnné jsme byli schopni přesně analyzovat vliv umělého elektronového akceptoru PPBQ na S-stavy u vyšších rostlin a porovnat s měřením a simulacemi deaktivace S-stavů u sinic. Výsledek ukázal na možnou existenci procesu relaxace nezávisle na účinku PPBQ u S3-stavu. Jako možný mechanismus relaxace jsme navrhli pomalou kooperaci mezi S3PSIIa a S3PSIIb u sinic a mezi S3PSIIa a S3,2,1PSIIb u vyšších rostlin.It is known that the photosynthesis is the process which is behind the breathable oxygenic atmosphere; the source of oxygen is photosynthetic dissociation of water molecules. A useful way to examine water splitting is flashes-induced measurement of the evolved oxygen. The short and intense flashes produce an oxygen oscillation with a periodicity of four. In order to describe and explain the oxygen oscillation, a kinetic modeling is one of the suitable methods which can be used. The explanation of the damping in the oxygen oscillation has been based on the kinetic parameters introduced by Bessel Kok. However, the meaning of these parameters was not clear because earlier kinetic models of water splitting were simplified. This thesis is therefore based on kinetic modeling and a description of water splitting. Initially, a new model of photosystem II (PSII) for simulation of the period-four oscillatory behavior of oxygen evolution (and chlorophyll fluorescence) was developed. An analysis of several outputs compared with experiments revealed that water splitting cannot be described in the PSII kinetic model without considering the initial phase of water splitting, the so-called intermediate S-states. These intermediate S-states, [SnYZox]-states (n = 0, 1, 2, 3) with rate constant of their formation kiSn ~ 1.5 × 106 s-1, were explained as a relaxation process associated with changes in the redox equilibrium P680+YZ ? P680YZox. This new model was the first step at simulating oxygen oscillation without the miss and double hits parameters, as defined by Bessel Kok. In order to uncover what is actually behind the double hits (oxygen evolution after the second flash), a new sophisticated model was developed. On the basis of this model it was suggested that it is very likely that the dimeric organization of the PSII leads to cooperation in the water splitting (suggested for cyanobacteria as an evolutionary mechanism at low light conditions. The cooperation was identified as a process which is behind the part of Kok parameter double hits if the sample is induced by single turnover flashes. Finally, in order to improve description of the dark decay processes related to water splitting, improved PSII model was developed. In this model, all the cofactors within the PSII were considered in one multi-state variable. An analysis of data from the cyanobacteria and higher plants treated or untreated by the artificial electron acceptor PPBQ revealed that a further decay mechanism, the relaxation, occurs in the S3-state, even in the PPBQ-free sample. The mechanism of the relaxation was explained by the involvement of slow pathways of cooperation between S3PSIIa and S3PSIIb in the cyanobacteria and between S3PSIIa and S3,2,1PSIIb in the higher plants.
