Cílem předložené diplomové práce je studium vlivu nanočástic stříbra na bakterie střevní mikroflóry. K testování byly vybrány dva standardní referenční bakteriální kmeny kolonizující trávicí trakt Lactobacillus acidophilus CCM 4832 a Escherichia coli CCM 3954. Bakterie byly zakoupeny z České sbírky mikroorganizmů (CCM). Nanočástice stříbra připravené metodou redukce diamminstříbrného komplexu D-maltózou v přítomnosti roztoku hydroxidu sodného bylo nutno vzhledem k jejich interakci se zvoleným MRS kultivačním médiem stabilizovat. Jako stabilizátory byly použity kasein, polyakrylová kyselina a želatina. Antibakteriální aktivita nestabilizovaného, stabilizovaného koloidního stříbra, stejně jako roztoku iontového stříbra (AgNO3) a roztoků použitých stabilizátorů byla testována standardní mikrodiluční metodou v MRS médiu a Mueller-Hinton médiu vedoucí k určení minimální inhibiční koncentrace (MIC) látky. Mikrodiluční metoda je podle celosvětově respektované organizace NCCLS (National Committee for Clinical Laboratory Standards) standardní metodou pro vyšetření MIC antibiotik u téměř všech druhů klinicky významných bakterií. Výsledky testování MIC v MRS kultivačním médiu ukázaly vyšší antibakteriální aktivitu stabilizovaných nanočástic stříbra vůči bakteriím druhu Escherichia coli patřící mezi podmíněné patogeny ve srovnání s tzv. přátelskými bakteriemi rodu Lactobacillus acidophilus.The aim of my thesis is to stady the influence of silver nanoparticles on the enteric baceria. Two standard reference bacterial strains colonizing the digestive tract Lactobacillus acidophilus CCM 4832 and Escherichia coli CCM 3954 were selected for testing. These bacteria were purchased from the Czech Collection of Microorganisms. Nanoparticles of silver were synthesized by the reduction of the complex cation [Ag(NH3)2]+ with D-maltose in the presence of sodium hydroxide solution. It was needed to stabilize them due to their interaction with the selected MRS nutrien broth. Casein, polyacrylic acid and gelatine were used as a stabilizers. Antibacterial activity of the modified and unmodified aqueous silver nanoparticles dispersions as well as solution of ionic silver (AgNO3) and solutions of the used modifiers was tested by a standard microdilution method in MRS nutrien broth and Mueller-Hinton nutrien broth to determinate the minimum inhibitory concentration (MIC) of the substances. The microdilution method is according to the word-wide respected organization NCCLS (National Committee for Clinical Laboratory Standards) the standard method for testing the minimun inhibitory concentration of antibiotics in majority of strains of clinical significant bacteria. The results of testing of MIC in MRS nutrien broth showed higher antibacterial activity of the modified aqueous silver nanoparticles dispersions against bacterial strain Escherichia coli belonging to the conditional pathogens in comparison with the so-called friendly bacteria Lactobacillus acidophilus.
