Náplní práce je charakterizace struktury a funkce některých proteinkinas v normálních netransformovaných buňkách a dále mechanismů jejich onkogenní aktivace. Vzhledem ke vzniku rezistence vůči aplikovaným terapeutikům jsou popsány některé inhibitory nových generací a naznačeny důvody pro testování kombinací inhibitorů proteinkinas s klasickými cytostatiky či jinými cílenými preparáty. Hlavním praktickým příspěvkem této práce je studium kombinovaného vlivu imatinibu (inhibitoru Bcr-Abl kinasy) a roskovitinu (CDK inhibitoru) na buněčnou linii K562. Jejich účinnost a především účinnost jejich kombinace bude sledována několika nezávislými metodami.This thesis is focused on structures and functions of selected protein kinases in normal and transformed cells as well as on mechanisms their oncogenic activation. Pharmacological inhibitors that have recently entered market however often induce specific resistance. Therefore, second generations of inhibitors are being developed and tested not only as a monotherapy, but also in combinations with other chemotherapeutics. Practical part of this thesis describes combined action of Bcr-Abl inhibitor imatinib and CDK inhibitor roscovitine in cell line K562. Effects of both compounds alone and especially in combination are characterized by several independent methods.
