V genomu cyanobakterie Nostoc 7120 byl identifikován gen hypotetické cytokinindehydrogenasy (CKX), enzymu ireverzibilně degradujícího rostlinné hormony cytokininy. Je známo, že cyanobakterie jsou považovány za předchůdce rostlinných chloroplastů a předpokládá se, že cytokininy mají na cyanobakterie podobný vliv jako na rostliny, co se týče vývoje a růstu. Cyanobakteriální gen NsCKX byl klonován z vektoru pQE 40 do vektoru pMAL-c4X a exprimován v Escherichia coli. Exprese byla detekována pomocí SDS-PAGE elektroforézy. Rekombinantní protein byl poté purifikován a také mutován pro získání enzymové aktivity. Ani v jednom případě však nebyla aktivita zaznamenána. Pro omezení možných chyb při skládání proteinu po jeho expresi byl mutovaný i nemutovaný gen NsCKX klonován do vektoru pMAL-p5X, který umožňuje expresi do periplasmy. Exprese cílového proteinu však nebyla detekována a enzymovou aktivitu se také nepodařilo zaznamenat. Podmínky exprese proto budou muset být optimalizovány. Otázka, zda identifikovaný gen skutečně kóduje jeden z důležitých enzymů účastnících se metabolismu cytokininů, tak zůstává stále nezodpovězena.A gene for a putative cytokinin dehydrogenase, an enzyme which degrades plant hormones cytokinins, was identified in a genome of cyanobacteria Nostoc 7120. It is generally agreed, that plant chloroplasts have developed from a cyanobacterial ancestor and that cytokinins may have similar function in development and growth in both cyanobacteria and plants. Cyanobacterial NsCKX gene was cloned from the pQE 40 vector into the pMAL-c4X vector and this recombinant vector was then expressed in Escherichia coli. Expression was detected by SDS-PAGE electrophoresis. Recombinant protein was then purified and also site-directed mutagenesis was performed to obtain enzyme activity. However, active protein was not obtained in any case. To reduce possible errors in protein folding after expression, wild-type and mutated genes were cloned into the pMAL-p5X vector that allows the expression into a periplasm. Neither expression of the target protein nor enzyme activity was detected. Expression conditions have to be further optimized. The question of whether the identified gene indeed encodes an important enzyme of cytokinin metabolism thus still remains unanswered.
