Diplomová práca je zameraná na sledovanie expresie bariérových génov v črevnej bunkovej línii Caco-2 po navodení zápalu inflamačnými faktormi lipopolysacharidom, tumor nekrotizujúcim faktrorom alfa a interleukínom 1 beta. V teoretickej časti bola popísaná problematika fungovania intestinálnej bariéry a idiopatických črevných ochorení. V experimentalnej časti bol pomocou metódy kvantitatívnej polymerázovej reťazovej reakcie v reálnom čase sledovaný vplyv zápalových faktorov a ich zmesi na expresiu bariérových génov tight junction proteínu 1, okludínu a cadherínu 1.This thesis focuses on the expression of barrier genes in the intestinal cell line Caco-2 after induction of inflammation by the inflammatory factors lipopolysaccharide, tumour necrosis factor alfa and interleukin 1 beta. In the theoretical part, the functioning of the intestinal barrier and idiopathic intestinal diseases were described. In the experimental part, the effect of inflammatory factors and their mixture on the expression of barrier genes tight junction protein 1, occludin, and cadherin 1 was observed using a real-time quantitative polymerase chain reaction method.
