Diplomová práce se zabývá vývojem analytických metod vhodných pro zjišťování přítomnosti fentanylu a jeho degradačního produktu norfentanylu ve směsích s dalšími opioidy (kokain, heroin a morfin). Teoretická část je zaměřena na voltametrickou a chromatografickou analýzu uvedených opioidů. V praktické části byly vyvinuty dvě analytické metody pro detekci fentanylu využívající adsorptivní cyklickou square-wave voltametrii (AdsCSWV) na dvou typech komerčních sítotiskových uhlíkových elektrod (typy C110 a 110CNT od firmy Metrohm DropSens). Testována byla opakovatelnost měření (jako relativní směrodatná odchylka) fentanylu (0,1 mmol/l) metodou AdsCSWV na elektrodách stejného typu, která představovala 7-8 % u elektrod typu 110CNT a 21 23 % u elektrod typu C110. Pro obě elektrody byly z regresních kalibračních přímek zjištěny meze detekce a meze stanovitelnosti fentanylu. Metoda AdsCSWV pro C110 elektrody byla úspěšně testována na směsných roztocích fentanylu s heroinem nebo kokainem. V chromatografické části práce byly nalezeny dva separační systémy, využívající CN kolonu a duální spektrofotometrickou a coulometrickou detekci, vhodné pro analýzu studovaných opioidů.The Master's thesis deals with the development of analytical methods suitable for the detection of fentanyl and its degradation product norfentanyl in their mixtures with other opioids (cocaine, heroin and morphine). The theoretical part is focused on voltammetric and chromatographic analysis of these opioids. In the practical part, two analytical methods were developed for the detection of fentanyl using adsorptive cyclic square-wave voltammetry (AdsCSWV) on two types of commercial screen-printed carbon electrodes (SPE, type C110 and 110CNT from Metrohm DropSens). The repeatability (as relative standard deviation) of the measurement of fentanyl (0,1 mmol/l) by AdsCSWV method was tested on the electrodes of the same type. It was 7-8 % for the 110CNT and 21-23 % for the C110 SPEs. For both electrodes, the limits of detection and the limits of quantification for fentanyl were determined from regression calibration lines. The AdsCSWV method for SPE C110 was successfully tested on mixed solutions of fentanyl with heroin or cocaine. In the chromatographic part, were found two separation systems with CN-column and dual spectrophotometric and coulometric detection suitable for analysis of studied opioids.
