Hlavním cílem této bakalářské práce bylo zjistit, zda jsou aniontové kanály zapojeny v růstu a vývoji rostlin Arabidopsis thaliana (L.) Heynh. a pokud ano, zda může být jejich aktivita ovlivňována světlem a prostřednictvím receptorů modrého světla, fototropinů. Teoretická část shrnuje dosavadní poznatky o fotomorfogenezi rostlin a fotoreceptorech, charakterizuje aniontové kanály a jejich inhibitory a popisuje vliv světla na aniontové kanály. V experimentální části byl použit genetický přístup, který spočíval v charakterizaci růstu fotomorfogenních mutantů phot1, phot2 a phot1/2 v přítomnosti kyseliny niflumové (NIF), inhibitoru aniontových kanálů. V podmínkách in vitro byl během experimentů sledován účinek vybraných koncentrací NIF v kultivačním médiu na růst rostlin a vývoj rostlin, a to v závislosti na světelných podmínkách (tma, modré a červené světlo). Výsledky experimentů naznačují, že aniontové kanály pozitivně ovlivňují růst rostliny a jejich aktivita může být regulována světlem prostřednictvím fotoreceptorů PHOT1 a/nebo PHOT2.The main aim of this bachelor thesis was to investigated whether anion channels are involved in the growth and development of Arabidopsis thaliana (L.) Heynh. plants and if so, whether their activity can be influenced by light and through blue light receptors, phototropins. The theoretical part summarizes the current knowledge on plant photomorphogenesis and photoreceptors, characterizes anion channels and their inhibitors, and describes the effect of light on anion channels. In the experimental part, a genetic approach was used to characterize the growth of phot1, phot2 and phot1/2 photomorphogenic mutants in the presence of niflumic acid (NIF), an anion channel inhibitor. Under in vitro conditions, the effect of selected concentrations of NIF in the culture medium on plant growth and development was monitored during experiments under light conditions (dark, blue and red light). The results of experiments suggest that anion channels are positively involved in plant growth and their activity can be regulated via photoreceptors PHOT1 and/or PHOT2.
