Tato práce se zabývá analýzou bodových mutací, které doprovázejí patogenezi vybraných hematologických onemocnění a slouží jako diagnostické markery u systemové mastocytózy (mutace D816V genu c-KIT) a vlasatobuněčné leukemie (mutace V600E genu BRAF) a/nebo představují slibné terapeutické cíle a potenciální markery reziduální nemoci (mutace genů IDH1/IDH2 u akutních myeloidních leukemií). Digitální PCR představuje časově a finančně nenáročnou molekulární techniku, která umožnuje při nízkém nároku na vstupní genetický materiál spolehlivě detekovat vybrané genové mutace i při jejich nízké alelické frekvenci. Experimentální část této práce byla provedena na 2 různých platformách dPCR, na systému Naica? (Stilla Technologies) a na QIAcuity (Qiagen). V práci byly shrnuty výsledky získané na základě detekce mutací IDH1/IDH2 a BRAF pomocí komerčně dostupných kitů (ID Solutions). Druhá část výsledků byla získána během optimalizace vlastních metod detekce vybraných mutací genů c-KIT a BRAF pomocí digitální PCR na systému QIAcuity. Určení bodových mutací pomocí digitální PCR vhodně doplní dostupné techniky analýzy bodových mutací založené na alelově specifické PCR amplifikaci a sekvenování. Výsledky této práce přispějí k zpřesnění analýzy bodových mutací c-KITD816V a BRAFV600E v Laboratoři molekulární biologie HOK FNOL.This diploma thesis focuses on the analysis of point mutations that associate with the pathogenesis of selected haematological disorders and serve as diagnostic markers in systemic mastocytosis (D816V mutation of c-KIT gene) and hairy cell leukemia (V600E mutation of BRAF gene) and/or represent promising therapeutic targets and potential markers of residual disease (IDH1/IDH2 gene mutations in acute myeloid leukemias). Digital PCR is a time-and cost-efficient molecular technique that enables reliable detection of selected gene mutations even at low allelic burden with low input genetic material requirements. The experimental part of this work was performed on 2 different dPCR platforms, the naica? system (Stilla technologies) and the QIAcuity system (Qiagen). The summarized results were obtained during the detection of IDH1/IDH2 and BRAF mutations using commercially available kits (ID solutions). The second part of the results was gained during the optimization of in-house methods for the detection of selected c-KIT and BRAF gene mutations using digital PCR on the QIAcuity system. Identification of point mutations by digital PCR will suitably complement available point mutation analysis techniques based on allele-specific PCR amplification and sequencing. The results of this work will contribute to the refinement of the analysis of point mutations c-KIT D816V and BRAF V600E point mutations in the Laboratory of Molecular Biology at the University Hospital Olomouc.
