Střevní mikrobiom má významný vliv na lidský organismus a je spojován s rozvojem celé řady onemocnění. Střevní mikrobiom ovlivňuje také metabolismus léčiv a tím potencionálně i výsledek farmakoterapie. Nejdůležitějšími biotransformačních enzymy I. fáze jsou cytochromy P450 (CYP), které jsou zodpovědné za biotransformaci 70-80 % všech klinicky užívaných léčiv. Cílem bakalářské práce bylo studium vlivu střevního mikrobiomu na enzymovou aktivitu vybraných CYP v mikrosomální frakci izolované z jater myších samců a samic. Pro posouzení vlivu mikrobiomu byly použity gnotobiotické myši s přesně definovaným složením střevního mikrobiomu. Ze získaných dat vyplývá, že střevního mikrobiom a kolonizace bezmikrobních myší probiotickou bakterií Escherichia coli Nissle 1917 (EcN) ovlivňuje aktivity vybraných CYP v játrech. Zároveň byly pozorovány rozdíly vlivu bezmikrobního stavu a kolonizace EcN u samců a samic. Vliv střevního mikrobiomu spolu s dalšími faktory (např. pohlaví) by tak neměl být opomíjen při hledání bezpečné a efektivní farmakoterapie.The gut microbiome influences significantly human health and predisposition to many diseases. The gut microbiome also influences drug metabolism and thus potentially the outcome of pharmacotherapy. The most important phase I biotransformation enzymes are cytochromes P450 (CYPs), which are responsible for the metabolism of 70-80 % of all clinically used drugs. The aim of the bachelor thesis was to study the influence of the gut microbiome on the enzyme activity of selected CYPs in the microsomal fraction isolated from the livers of male and female mice. To assess the effect of microbiome, gnotobiotic mice with a precisely defined composition of the intestinal microbiota were used. The obtained data has shown that the gut microbiome and the colonization of germ- free mice with the probiotic bacteria Escherichia coli Nissle 1917 (EcN) influence the activities of selected CYPs in the liver. Further, differences in the effect of the germ-free state and EcN colonization were observed in male and female mice. Therefore, the influence of the gut microbiome along with other factors (e. g. gender) should not be neglected in the search for safe and effective pharmacotherapy.
