Zmp1 je enzym, jenž podporuje šíření Mycobacterium tuberculosis v těle hostitele a vznik granulomu chránícího infikované makrofágy před útoky imunitního systému. Tuberkulóza je závažné onemocnění, kterým se každoročně nakazí 7,5 milionu lidí. Léčbu komplikuje narůstající počet mykobakteriálních kmenů rezistentních vůči současným antituberkulotikům. V současné době je vyvíjena snaha syntetizovat nové látky zabraňující působení M. tuberculosis. Tato práce je zaměřena na indolové hydroxamáty inhibující aktivitu enzymu Zmp1. Účinnost jednotlivých látek na inhibici mykobakteriálního enzymu Zmp1 je zjištěna pomocí MALDI ToF hmotnostní spektrometrie.Zmp1 is an enzyme that promotes the spread of Mycobacterium tuberculosis in the host body and the formation of a granuloma that protects infected macrophages from immune attacks. Tuberculosis is a serious disease that infects 7.5 million people each year. Treatment is complicated by the increasing number of mycobacterial strains resistant to current antituberculosis drugs. Efforts are currently underway to synthesise new agents to inhibit M. tuberculosis. This bachelor thesis focuses on indole-hydroxamates that inhibit the activity of the Zmp1 enzyme. The efficacy of each compound to inhibit the mycobacterial Zmp1 enzyme is analyzed by MALDI ToF mass spectrometry.
