Předložená bakalářská práce je zaměřena na modernizaci postupů izolace a stanovení vybraných fytoekdysteroidů v rostlinných pletivech se zaměřením na profilování jak bioaktivních látek s fytoekdysteroidní strukturou, tak jejich biosyntetických prekurzorů. Cílem této bakalářské práce bylo zjistit, zda chemický charakter prekurzorů a produktů biosyntézy umožňuje společný postup extrakce a izolace a tento postup dále optimalizovat. Pro přesnou kvantifikaci dále použít jak již zavedenou metodiku na bázi LC-MS, tak pro biosyntetický prekurzor zavést zcela novou metodiku využívající stejný analytický přístup. Teoretická část této bakalářské práce se tak zaměřuje zejména na biosyntézu fytoekdysteroidů a experimentální část práce je členěna na tři části: 1. část zahrnující vývoj a optimalizaci extrakce a purifikace fytoekdysteroidů a lathosterolu - jejich biosyntetického prekurzoru v laskavcovitých (Amaranthaceae); 2. část zahrnující optimalizaci navážky vybraného pletiva z čeledi laskavcovitých (špenátu setého Spinacia oleracea) a 3. část je věnována vývoji LC-MS metody pro stanovení lathosterolu metodou izotopového zřeďování. Práci uzavírá kapitola výsledky a závěr.The submitted bachelor's thesis is focused on the modernization of isolation procedures and the determination of selected phytoecdysteroids (PEs) in plant tissues with a focus on profiling both bioactive substances with a PE structure and their biosynthetic precursors. The aim of this bachelor's thesis was to find out whether the chemical nature of the precursors and products of biosynthesis allows for a common procedure of extraction and isolation and to further optimize this procedure. The next goal was to introduce completely new methodology for quantification of their biosynthetic precursor. The theoretical part of this bachelor thesis focuses mainly on the biosynthesis of PEs and their distribution. The experimental part of the work is divided into three parts: the first part includes the development and optimization of extraction and purification of PEs and lathosterol - their biosynthetic precursor in Amaranthaceae; The second part includes the optimization of the weight of the selected tissue from the Amaranthaceae family (Spinacia oleracea) and the third part is devoted to the development of the LC-MS method for the determination of lathosterol by the isotopic dilution method.
