Tato práce pojednává o studiu auxin-interagujících proteinů v huseníčku rolním (Arabidopsis thaliana) vybranými proteomickými metodami. Proteiny byly izolovány pomocí nativní extrakce, která hraje klíčovou roli zejména při zachování přirozené konformace proteinů. Význam nativní extrakce byl uplatněn v dalším kroku experimentu, kterým byla afinitní chromatografie. Nativní forma proteinů byla důležitým faktorem pro přirozenou afinitu proteinů k ligandům. Eluované proteiny byly frakcionovány a následně identifikovány tandemovou hmotnostní spektrometrií (ESI-QTOF). Získané informace mohou posloužit k objasnění role auxin-interagujících proteinů v mechanismu účinku auxinu na rostlinu a zároveň doplnit znalosti o vlastnostech auxinu a jeho fyziologických účincích.The thesis deals with the study of auxin-binding proteins in Arabidopsis thaliana using the chosen proteomic methods. The proteins were isolated using the native protein extraction which plays a key role especially while maintaining the natural conformations of proteins. The significance of native protein extraction was utilized in the next step of the experiment which was affinity chromatography. The native state of proteins was an important factor for natural affinity of proteins for ligands. The eluted proteins were fractionated and then identified using tandem mass spectrometry (ESI-QTOF). The gathered information can serve to clarify the role of auxin-binding proteins in the mechanism of effect of auxin on plant and at the same time, complement the knowledge about the characteristics of auxin and its physiological effects.
