Diplomová práce se zabývá problematikou analýzy biologicky aktivních látek pomocí molekul aptamerů. Ty by mohly mít potenciál nahradit při analýze v současnosti používané protilátky nebo enzymy. Aptamery ve spojení s vhodnými převodníky fyzikálně-chemického signálu tvoří tzv. aptasenzory. Jako převodníky jsou často využívané nanomateriály díky svým specifickým vlastnostem. V této práci byly jako převodníky signálu využity senzory obsahující nanovrstvu zlata. Jako modelová interakce aptasenzoru sloužila hybridizace oligonukleotidů. Oligonukleotid sloužící jako aptamer má strukturu hairpinu, a při interakci s komplementárním oligonukleotidem dochází k rozvětvení jeho struktury. Interakce byla charakterizována pomocí metod povrchové plasmonové rezonance, povrchem zesílené Ramanovy spektroskopie a metody kombinující povrchovou plasmonovou resonanci a elektrochemii.This diploma thesis focuses on the analysis of biologically active compounds using aptamer molecules. These molecules could have the potential to replace currently used antibodies or enzymes in the analysis. Aptasensors are formed by aptamers in combination with suitable physicochemical signal transducers. Due to their specific properties, nanomaterials are often used as transducers. In this work, sensors with a gold nanolayer were used as signal transducers. Oligonucleotide hybridization was used as a model for aptasensor interaction. The oligonucleotide with hairpin structure was utilized as an aptamer. The hairpin structure unfolds as a results of the interaction with the other oligonucleotide sequence. The interaction was characterized by surface plasmon resonance, surface-enhanced Raman spectroscopy and by a method combining surface plasmon resonance and electrochemistry.
