DNA-proteinové crosslinky představují pro buňky vysoce toxické poškození v podobě kovalentního spojení proteinů s řetězcem DNA. Pokud nejsou včas opraveny, může docházet k mutacím a buněčné smrti. Naštěstí si buňky vytvořily efektivní cesty, kterými dokážou obnovit původní strukturu genetického materiálu a zachovat tak integritu genomu. Pro identifikaci genů, které hrají roli v opravách DNA-proteinových crosslinků, byl ve vědecké skupině doc. Aleše Pečinky navržen dopředný genetický screen, který využívá EMS-mutagenizované populace rostlin huseníčku rolního (Arabidopsis thaliana). K indukci DNA-proteinových crosslinků sloužil zebularin - cytidinový analog, jehož schopností je zachytávat enzym METYLTRANSFERÁZU 1 na DNA a vytvářet tak sterické zábrany pro jiné enzymy spjaté s DNA. Rostliny, ve kterých vlivem mutace došlo k inaktivaci kauzálního genu, vykazovaly díky ošetření zebularinem výraznou redukci kořene. Pomocí těchto postupů byly nově objeveny kandidátní linie 21B-5 a 21B-30, k analýzám charakterizujícím efekt mutace však v rámci experimentální části bakalářské práce sloužila pouze linie 21B-5.DNA-protein crosslinks represent highly toxic damage to cells in the form of a covalent linkage of proteins with a DNA strand. If not properly repaired, DNA-protein crosslinks can lead to mutation and cell death. Fortunately, cells have evolved effective DNA repair pathways to restore the original structure of genetic material and thus maintain their genomic integrity. To identify genes that play a role in DNA-protein crosslink repair, the research group of Doc. Aleš Pečinka designed a forward-directed genetic screen that uses EMS-mutagenized populations of the thale cress (Arabidopsis thaliana). Zebularine - a cytidine analog was used to induce DNA-protein crosslinks, because of its ability to covalently trap METHYLTRANSFERASE 1 on the DNA and thus create steric hindrance to other DNA-related enzymes. Plants in which the causal gene was inactivated by the mutation showed significant root reduction after the zebularine treatment. This allowed me to find candidate lines 21B-5 and 21B-30, but only line 21B-5 was used for analysis identifying and also characterizing the causal mutation as a factor involved in DNA-protein crosslink repair.
