Tato bakalářská práce se zabývá proteinem vitellogeninem. V teoretické části popisuje jeho vlastnosti, strukturu, biosyntézu a přínosné funkce v imunitě, antioxidační ochraně či dlouhověkosti u včely medonosné. Pozornost je věnována také popisu metod, sloužících k analýze vitellogeninu. V praktické části byla pomocí Bradfordovy metody zjištěna celková koncentrace proteinů včelí hemolymfy, které byly následně separovány pomocí gelové elektroforézy. Metodou Western blot byl ve vzorku hemolymfy detekován vitellogenin. Také byla provedena bioinformatická analýza, pomocí níž se podařilo najít u jiných druhů včel podobné sekvence proteinů. Dále byla predikována místa, kde může v sekvenci tohoto proteinu docházet k posttranslačním modifikacím. S využitím homologního modelování byly vytvořeny modely domén vitellogeninu a jemu podobných proteinů vg-like-A, -B a -C.This bachelor thesis deals with the protein vitellogenin. In the theoretical part it describes its properties, structure, biosynthesis and beneficial functions in immunity, antioxidant protection or longevity in the honey bee. Attention is also paid to the description of methods used for vitellogenin analysis. In the practical part, the total concentration of honey bee hemolymph proteins was determined using the Bradford method and these proteins were subsequently separated by gel electrophoresis. The Western blot method was used to detect vitellogenin in the hemolymph sample. Bioinformatic analysis was also performed and similar protein sequences were found in other bee species. Furthermore, the sites where post-translational modifications may occur in the sequence of this protein were predicted. Finally, models of the domains of vitellogenin, vg-like-A, -B and -C proteins were generated using homology modelling.
