Tato diplomová práce se zabývá proteomickou analýzou vzorků cervikálního hlenu s ohledem na potenciální identifikaci možných biomarkerů receptivity endometria. Nízká úspěšnost In vitro fertilizace je velmi známým a častým problémem celého procesu. Nejen, že má negativní vliv na budoucí rodiče, ale také výrazně zvyšuje finanční náročnost léčby. V této práci je uveden postup necílené proteomické analýzy s bioinformatickým zpracováním dat. V rámci analyzovaných vzorků došlo k vyčlenění skupiny zahrnující pouze těhotné pacientky. U těchto vzorků byly identifikovány specifické proteiny pro různé fáze ovulace: Annexin A1 a Apolipoprotein II (pre-ovulační fáze), Mucin-5AC (ovulační fáze) a Keratin I (post-ovulační fáze).This diploma thesis deals with the proteomic analysis of cervical mucus samples and the potential identification of possible biomarkers of endometrial receptivity. Low success rate of In vitro fertilization is a well-known and very common problem of the whole process. Not only that it has a negative effect on both future parents, but it also significantly increases the financial demands of the treatment. The non-targeted proteomic workflow with bioinformatic data processing is presented in this thesis. Within the analysed samples a group containing only pregnant patients was separated. And specific proteins for the different phases of ovulation were identified in these samples: Annexin A1, Apolipoprotein II (pre-ovulation phase), Mucin-5AC (ovulation phase) and Keratin I (post-ovulation phase).
