Tato bakalářská práce je věnována N-acetylaminotransferase (NATA) z mechu Physcomitrella patens. V teoretické části je popsán metabolismus polyaminů, jejich syntéza a degradace se zaměřením na acetylaci a známé enzymy u rostlin a savců. V experimentální části byl klonován gen PpNATA do expresního vektoru pET-28a. Expresí genu v buňkách E.coli byl získán rekombinantní protein obsahující histidinovou kotvu umožňující purifikaci afinitní chromatografií. Pomocí metody SDS-PAGE byla ověřena produkce proteinu, čistota a metodou nano-diferenciální skenovací fluorimetrie bylo ověřena stabilita proteinu a stabilizace vazbou na acetyl-CoA. Pomocí bioinformatických postupů byla predikována sekundární a terciární struktura proteinu na základě dostupných struktur savčí N-acetylaminotransferasy a dále bylo analyzováno vazebné místo pro vazbu acetyl-CoA v modelu PpNATA.This bachelor thesis is focused on the N-acetyl aminotransferase (NATA) from the moss Physcomitrella patens. The theoretical part describes the metabolism of polyamines, their synthesis and degradation with a focus on acetylation and known enzymes in plants and mammals. In the experimental part, the PpNATA gene was cloned into the expression vector pET-28a. The gene was expressed in E. coli cells yielding a recombinant protein comprising a histidine-tag allowing the affinity chromatography purification. The protein production and purity were verified by SDS-PAGE, and protein stability and the effect of acetyl-CoA binding was analyzed by the nano-differential scanning fluorimetry. Bioinformatic approaches were used to predict the secondary and tertiary structure of the protein was predicted based on the available structural data of N-acetyl aminotransferases, and the binding site for acetyl-CoA in the PpNATA model was further analyzed.
