Cílem práce bylo vyvinout metodu detekce rezistence bakterií, která by byla rychlejší než dosud využívané testy. Pro detekci enzymu produkovaného bakteriemi byly použity kompozity s polystyrenovými mikročásticemi s magnetickým jádrem a adsorbovanými nanočásticemi zlata nebo stříbra na povrchu, které byly redukovány buď kyselinou askorbovou nebo dopaminem. Měření probíhalo pomocí povrchem zesílené Ramanovy spektroskopie. Za využití kompozitů bylo možné pomocí Ramanovy spektroskopie detekovat přítomnost ampicilinu jako modelového -laktamového antibiotika. Přidáním enzymu docházelo k vymizení charakteristických píků antibiotika, nicméně zároveň nedocházelo k detekci produktu degradace působením -laktamázy.The aim of this thesis was to develop faster detection method of bacterial resistance than now used tests. For enzymatic detection produced by bacterium were used composites of polystyrene microparticles with magnetic core and adsorbed gold or silver nanoparticles on surface, reduced by ascorbic acid or dopamine. The measurement was done using surface-enhanced Raman spectroscopy. We were able to measure presence of ampicillin as our model -lactam antibiotic with the use of composites. Addition of enzyme resulted in disappearing of characteristics peaks of ampicillin but at the same time no product of the degradation by influence of -lactamase was detected.
