Fenomén známý jako ultra slabá emise fotonů (UPE) pochází ze všech živých organismů. Jedná se o slabou chemiluminiscenci, která je detekována od ultrafialové po blízkou infračervenou (IČ) oblast spektra. V této diplomové práci jsme se zaměřili na vyzařování excitační energie singletního kyslíku (1O2), která je detekována v červené a blízké IČ oblasti spektra. Singletní kyslík vzniká buď prostřednictvím Russellova mechanismu, tedy rozpadem tetroxidu (ROOOOR), nebo přenosem excitační energie z excitované molekuly na molekulu kyslíku v základním stavu. Excitační energie 1O2 je vyzařována ve formě monomolové nebo dimolové emise. V blízké IČ oblasti spektra při vlnové délce 1270 nm je detekována monomolová emise. Dimolová emise pochází ze dvou molekul 1O2 a je detekována v červené oblasti spektra při vlnových délkách 634 nm a 703 nm. V této diplomové práci jsme se zabývali tvorbou organických hydroperoxidů (ROOH) z chemického systému - hovězí sérum albumin. K albuminu byly přidány exogenní reaktivní formy kyslíku (ROS), které nastartovaly řetězovou reakci, která vedla až ke tvorbě 1O2. Následně jsme detekovali monomolovou a dimolovou emisi ze 1O2. Pro detekci jsme použili dva fotonásobiče a elektronovou paramagnetickou rezonanci (EPR).The phenomenon known as ultra-weak photon emission (UPE) is common for all living organisms. It is a weak chemiluminescence that is detected in the range from the ultraviolet to the near infrared (IČ) region of the spectrum. In this diploma thesis we focused on the emission of excitation energy of singlet oxygen (1O2), which is detected in the red and near IČ regions of the spectrum. Singlet oxygen is formed either through the Russell Mechanism, i.e. the decay of tetroxide (ROOOOR), or by the transfer of excitation energy from the excited molecule to the oxygen ground state molecule. The excitation energy of 1O2 is radiated in the form of monomolar or dimolar emission. Monomolar emission is detected in the near IČ region of the spectrum at the wavelength of 1270 nm. Dimolar emission comes from two 1O2 molecules and it is detected in the red region of the spectrum at 634 nm and 703 nm. In this diploma thesis we dealt with the formation of organic hydroperoxides (ROOH) from the chemical system - bovine serum albumin. Exogenous reactive oxygen species (ROS) were added to the albumin, which started a chain reaction that led to the formation of 1O2. Subsequently, we detected the monomolar and dimolar emission from 1O2. We used two photomultipliers and electron paramagnetic resonance (EPR) to detect the emission.
