Tato bakalářská práce se zabývá detekcí stopových množství oligonukleotidů, čehož se dá využít například u detekce virů v klinických vzorcích. V současnosti se k detekci nukleových kyselin využívá dvou přístupů: množení oligonukleotidů (například pomocí techniky PCR) nebo molekulární hybridizace. Na základě hybridizace mohou být k detekci využity molekuly aptamerů. Ty spolu s vhodnými nanomateriály tvoří tzv. aptasenzory. Cílem práce byl vývoj aptasenzoru na bázi povrchem zesílené Ramanovy spektroskopie (SERS), ta by měla být schopna detekovat i vzorky o velmi nízkých koncentracích. Jako model interakce aptameru s cílovou molekulou zde sloužily 2 sekvence vůči sobě komplementárních oligonukleotidů, kdy jeden z oligonukleotidů má strukturu hairpinu a při interakci dochází k rozevření struktury. Jako substrát zde byly využity čipy obsahující stříbrné nanostruktury.This bachelor thesis focuses on the detection of trace amounts of oligonucleotides. For example, this method can be used in detection of virus in clinical samples. Currently, two approaches are used to detect nucleic acids: oligonucleotide propagation (e.g., by PCR) or molecular hybridization. In the molecular hybridization approach, aptamer molecules are being used for detection when combined with suitable nanomaterials. The aim of this thesis was to develop an aptasensor based on surface-enhanced Raman spectroscopy. This method should be able to detect substances in samples of very low concentrations. Two sequences of oligonucleotides complementary to each other were used as a model of the interaction of aptamer with the target molecule. One of the oligonucleotides has a hairpin structure which unfolds as a result of an interaction with the other oligonucleotide. Wafers containing silver nanostructures were used as a substrate.
