Kyselina gamma-aminomáselná (GABA) je neproteinogenní aminokyselina, k jejíž akumulaci v rostlinách dochází v odpovědi na řadu biotických a abiotických stresů. Cílem této práce bylo prozkoumat úlohu GABA metabolismu v růstu A. thaliana za solného stresu. Knockoutované rostliny s T-DNA inzercí v devíti genech metabolismu GABA byly selektovány na plotnách s antibiotikem a poté genotypovány pomocí PCR. Homozygotní rostliny byly získány v případě sedmi genů. Růst a další fenotypové vlastnosti těchto homozygotních linií byly posouzeny za kontrolních podmínek a podmínek solného stresu s využitím fenotypování. Mezi liniemi byly identifikovány fenotypové odlišnosti. Dle našich výsledků se geny GAD1, GAD4, GABA-T a GLYR2 podílely na růstu rostlin, zatímco gad3 mutanti vykazovali fenotyp odlišný od ostatních mutantních linií. V budoucnosti by mohl být proveden další experiment s využitím různých koncentrací soli.Gamma-aminobutyric acid (GABA) is a non-proteinogenic amino acid which, in plants, rapidly accumulates in response to a variety of biotic and abiotic stresses. The aim of this thesis was to investigate the role of GABA metabolism in the growth of A. thaliana under salt stress. Knockout plants with T-DNA insertion in nine GABA metabolism genes were selected on plates with antibiotics and then genotyped by PCR. Homozygous plants were successfully obtained for seven genes. The growth and other phenotypic related traits of these homozygous lines were evaluated under control and salt stress conditions using phenotyping approaches. Phenotypical differences among the lines were identified. According to our results, GAD1, GAD4, GABA-T and GLYR2 genes were involved in plant growth, while the gad3 mutants showed a phenotype different from other mutant lines. In the future, another experiment using different concentrations of salt could be performed.
