V rámci předkládané diplomové práce byla ověřená exprese chimérních genů obsahujících sekvence kódující fragmenty virálních proteinů: G-protein viru jarní virémie kaprů a membránový protein ORF 149 viru koiherpesvirózy. Transgeny vloženy do vektoru pGWB17 byly využity na transientní transformaci listů tabáku Nicothiana benthamiana infikovaných bakteriální kulturou Agrobacterium tumefaciens GV3101. Pro detekci rekombinantích proteinů byla využita fluorescenční mikroskopie a metoda Western blot. Za účelem testováni exprese transformačních konstruktů vložených do vektoru pBRACT214 byla vykonána transientní transformace buněk listů ječmene setého (Hordeum vulgare cv. Golden Promise) využitím balistické metody. Následně, za účelem propagace transgenních rostlin ječmene byla vykonána stabilní transformace nezralých ječmenných embryí bakteriální kulturou Agrobacterium tumefaciens AGL1, a to transformačními konstrukty pro expresi G-proteinu. Rekombinantní protein byl v nediferencovaných buňkách kalusů detekován pomocí fluorescenční mikroskopie.In this diploma thesis there was verified the expression of chimeric genes containing sequences encoding fragments of viral proteins: G-protein of spring carp viraemia virus and membrane protein ORF 149 of koiherpesvirus. The transgenes inserted into the pGWB17 vector were used to transiently transform Nicothiana benthamiana tobacco leaves, infected with Agrobacterium tumefaciens GV3101 bacterial culture. Fluorescence microscopy and Western blotting were used to detect recombinant proteins. In order to test the expression of transformation constructs inserted into the pBRACT214 vector, transient transformation of barley leaf cells (Hordeum vulgare cv. Golden Promise) was performed by using the ballistic method. Subsequently, in order to propagate the transgenic barley plants, a stable transformation of immature barley embryos was performed with a bacterial culture of Agrobacterium tumefaciens AGL1, using transformation constructs for G-protein expression. Recombinant protein was detected in undifferentiated callus cells by fluorescence microscopy.
