V této bakalářské práci jsem se zabývala hledáním polymorfních mikrosatelitů u pelikána bílého (Pelecanus onocrotalus) pomocí cross species PCR amplifikace. V teoretické části jsem popsala řád pelikáni, čeleď pelikánovití a pelikána bílého jako testovaný druh, včetně jeho obtížného zařazení do systému, který je stále předmětem studií. Dále je zde uvedena charakteristika mikrosatelitů, jejich dělení, mutace a význam. Na konci jsou popsány de novo a cross-species mikrosatelity určené pro testování na pelikánu bílém. V praktické části jsem testovala polymorfismus mikrosatelitních lokusů na genomické DNA 6 nepříbuzných jedinců pelikána bílého užitím 213 párů primerů, z nichž 207 bylo izolováno z řádu trubkonosí, 5 z řádu dlouhokřídlí a 1 pocházel od zástupce z řádu pěvci. Tyto páry primerů poskytly u zástupců z řádu trubkonosí polymorfní produkt, proto byly do testování zahrnuty. Cross-species PCR amplifikací jsem detekovala 46 párů primerů poskytujících 47 polymorfních produktů, protože pár primerů Dc5 poskytuje produkty dva. Ze zbytku bylo 163 monomorfních a čtyři ani při teplotě annealingu 44 °C neposkytly žádný produkt.In this bachelor thesis, I focused on finding polymorphic microsatellite loci of the Great White Pelican using cross-species PCR amplification. In the theoretical part, I described the order Pelecaniformes, family Pelecanidae and the Great White Pelican as a tested species. I also mentioned the problems with its classification which still makes this species a study object. Furthermore, there is a characteristic of microsatellites, their classification, mutation and utilization. At the end of this part, I described de novo and cross-species pairs of primers that were derived from the original species and used for testing. In the practical part, I tested 213 microsatellite loci using appropriate pairs of primers. Two hundred and seven pairs of primers were designed for the Procellariiformes species, another six pairs of primers were originally designed for the Charadriiformes and Passeriformes. Cross species PCR amplification along with these pairs of primers provided a polymorphic product to the representatives of Procellariiformes and because of that were included into the experiment. Using cross-species PCR amplification, I detected 46 polymorphic loci with 47 polymorphic PCR products. The reason is that one pair of primers amplified two regions of polymorphism. 163 microsatellite loci were monomorphic and 4 microsatellite loci did not provide any product.
