Studium diferenciace buněk je důležitým nástrojem k poznávání nových mechanismů metabolismu různých látek a také k vývoji nových metod v léčbě onkologických onemocnění. V této bakalářské práci byla experimentálně studována role enzymu solubilní epoxidhydrolázy v diferenciaci střevních buněk in vitro na nádorové buněčné linii HT-29. Buňky v různých stupních diferenciace byly inkubovány s inhibitorem solubilní epoxidhydrolázy a jejich proliferační aktivita byla poté měřena spektrofotometricky. Dále se měřila relativní exprese markerů diferenciace intestinální alkalické fosfatázy a villinu po ovlivnění inhibitorem pomocí In Cell ELISy. Dalším krokem bylo semikvantitativní hodnocení exprese solubilní epoxidhydrolázy, která se hodnotila u vzorků normální a nádorové tkáně od pacientů s kolorektálním karcinomem. Po použití inhibitoru solubilní epoxidhydrolázy bylo pozorováno snížení proliferace u buněk nediferencovaných a během diferenciace. Relativní exprese markerů diferenciace byla u těchto buněk také snížena. V rámci semikvantitativního hodnocení pak bylo prokázáno snížení exprese solubilní epoxidhydrolázy v nádorové tkáni málo diferencovaných nádorů oproti zdravé tkáni u 6 z 8 pacientů. Může se tak spekulovat o tom, že solubilní epoxidhydroláza hraje zásadní roli při diferenciaci střevních buněk, a že by se jejích účinků dalo využít pro lokální ovlivnění nediferencovaných nádorových buněk, které představují pro pacienta větší nebezpečí a tyto nádory jsou více agresivní.Understanding of cell differentiation is an important tool for recognizing new mechanisms of metabolism of various substances and for developing of potential new treatments of oncological diseases. In this bachelor thesis, the potential role of soluble epoxide hydrolase (sEH) in cell differentiation of intestinal cells in vitro was studied using the tumor cell line HT-29. The cells in different stages of differentiation were incubated with the inhibitor of soluble epoxide hydrolase and their proliferation activity was measured by spectrophotometry. Then the relative expression of differentation markers, intestinal alkaline phosphatase and villin, was measured by the In Cell ELISA method. Moreover, the expression of soluble epoxide hydrolase in normal and tumor tissue samples was detected immunohistochemically. After the inhibitor treatment the decrease in proliferation of the undifferentiated and differentiating cells was observed. Similarly, relative expression of intestinal alkaline phosphatase and villin was lower in undifferentiated and differentiating cells after sEH inhibitor treatment in comparison to control cells. Moreover, the statistically significant decrease in sEH expression in tumor tissue in comparison to normal tissue was observed in 6/8 patients with undifferentiated colorectal carcinoma. It can be speculated that soluble epoxide hydrolase could play essential role during intestinal cells differentiation and these effects could be used for local treatment of undifferentiated tumor cells, which represent higher risk for patients.
